酶法测定黄精含片中黄精多糖的研究

摘要 目的建立排除黄精片剂中辅料干扰多糖含量测定的方法。方法采用苯酚-硫酸比色法测定黄精多糖含量,糖化酶水解麦芽糊精,80%乙醇溶液进行醇沉分离,考察反应温度、p H、酶与底物比例及反应时间对酶解反应的影响,验证糖化酶能否水解黄精多糖。结果反应温度58℃、pH4.5、酶与底物比例1:5、反应时间28 min时麦芽糊精完全水解,80%醇沉后在490 nm处测定无明显吸收。采用酶水解法可以准确测定黄精片剂中多糖的含量,测定结果与加入量无差异(P>0.05)。结论糖化酶水解麦芽糊精,不水解黄精多糖,对黄精多糖含量的测定不存在干扰,可用于片剂中多糖含量测定。 Objective To establish a method that can remove the interference of production auxiliary materials on the content determination of polysaccharides that in solomonseal tablet. Methods The Polygonatum sibiricum polysaccharides content was detected by the phenol-sulfuric acid method; maltodextrin was treated with glucoamylase hydrolysis and then the Polygonatum sibiricum polysaccharides was separated by using ethanol precipitation method; the effect of temperature, p H, ratio of enzyme to substrate and hydrolysis time on hydrolysis effect was investigated and research whether the glucoamylase showed hydrolysis effects on polygonatum sibiricum polysaccharides. Results The maltodextrin was completely hydrolysised under these conditions: temperature 58℃, pH 4.5, ratio of enzyme to substrate 1 : 5 and hydrolysis time 28 min, the enzymatic hydrolysate showed no adsorption for 490 nm light after the ethanol precipitation reaction. The polygonatum sibiricum polysaccharides content that in solomanseal tablet can be determinated accurately by using glucoamylase hydrolysis method, the measurement result showed no significant difference with the addition amount(P 〉0.05). Conclusion Glucoamylase can hydrolysis maltodextrin but have no effect on polygonatum sibiricum polysaccharides, glucoamylase hydrolysis had no interference on the content determination of Polygonatum sibiricum polysaccharides, can be a reference for the content determination of polysaccharides that in tablet.
