摘要
目的研究尘肺病患者外周血细胞炎症相关基因的表达变化,探讨差异表达基因作为尘肺病指示基因的可能性。方法采集尘肺病患者及健康对照外周血样,提取总RNA并逆转录后,应用实时定量PCR阵列技术分析病例及对照外周血细胞中炎症相关基因表达情况,筛选差异表达基因。并对筛选出的部分差异表达基因进行初步验证。结果从84个炎症相关基因中筛选到11个差异表达基因(8个上调、3个下调)。对其中IL-6、IRF2及PTGS2基因的初步验证结果显示:尘肺病组IL-6 mRNA表达水平(0.638±0.376)明显高于对照组(0.492±0.366,P<0.05);尘肺病组IRF2及PTGS2 mRNA表达水平(1.655±0.734,2.769±2.000)与对照组(1.425±0.496,2.466±1.326)差异无统计学意义(P>0.05)。尘肺病组的IL-6 mRNA表达水平明显高于不吸烟对照组(P<0.05),不同病种、分期及脱尘时间对其表达没有影响(P>0.05)。结论实时定量PCR阵列是筛选尘肺病差异表达基因的有力工具,IL-6可能参与尘肺病的发生。
Objective To study the expression changes of inflammation - related genes in peripheral blood of pneumoconiosis patients, and explore the possibility of the screened genes as the pneumoconiosis indicators. Methods Peripheral blood samples of pneumoconiosis patients and the controls were collected, after the total RNA of the blood cells were extracted and reverse transcribed, and differentially expressed genes associated with inflammation between pneumoconiosis patients and controls were performed and screened by real - time quantitative PCR array. And the differential expression genes of the selected genes were preliminarily verified. Results The expression of 11 out of 84 genes was significantly altered in the pneumoconiosis patients compared with the control. Among theses 11 genes, 8 genes were up - regulated and 3 were down - regulated. Preliminarily ver- ified results indicated that the expression level of IL- 6 mRNA(0. 638 + 0. 376) in the pneumoconiosis group was significantly higher than that in the control group (0.492 + 0. 366, P 〈 0.05 ). There was no significant difference between IRF2 and PTGS2 mRNA expression levels ( 1. 655 + 0. 734 ,2. 769 + 2. 000 ) of the pneumoconiosis group and the control group ( 1. 425 + 0. 496, 2. 466 + 1. 326) ( P 〉 0.05 ). The expression level of mRNA 1L - 6 in the pneumoconiosis group was significantly higher than that in the control group ( P 〈 0.05 ). The expression of different diseases, stages and dust removal time had no effect on the ex- pression (P 〉 0.05 ). Conclusion Real -time quantitative PCR array is a powerful tool for screening the differential expres- sion genes of pneumoconiosis, and IL - 6 was involved in occurrence of pneumoconiosis.
