摘要
目的对比研究18α-甘草酸与18β-甘草酸对免疫性肝损伤小鼠的保护作用,并筛选二者联合用药的最佳配伍比例。方法采用刀豆蛋白A(Con A)诱导小鼠免疫性肝损伤模型,观察18α-和18β-甘草酸高、中、低剂量组,以及二者联合用药不同配伍比例组对肝损伤小鼠的丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、一氧化氮(NO)活性及其各脏器指数的影响,并检测肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)的含量,同时筛选二者联合用药的最佳配伍比例。结果与模型组比较,18α-和18β-甘草酸组均可降低小鼠血清中AST及肝匀浆中AST、ALT、NO活力,降低肝匀浆中TNF-α、IFN-γ的含量,改善小鼠肝脏、胸腺指数;比较二者对免疫性肝损伤小鼠的保护作用,差异无统计学意义;二者联合用药的最佳配伍比例为1∶4,其对肝匀浆中AST、ALT、NO水平的影响优于单用18α-、18β-甘草酸治疗组。结论 18α-和18β-甘草酸对小鼠免疫性肝损伤均有保护作用,二者联合用药的最佳配伍比例为1∶4,其对转氨酶及NO水平的影响优于单用18α-或18β-甘草酸。
Objective To compare the protective effect of 18α-and 18β-glycyrrhizic acid on immunological liver injury in mice and to study the compatibility proportion of 18α- and 18β-glycyrrhizic acid. Methods The mouse model of immunological liver injury was induced by concanavalin A(ConA). And then we observed the effect of high-, middle- and low-dose 18α- and 18β-glycyrrhizic acid as well as their combination at different compatibility proportions on activities of alanine aminotransferase(ALT), aspartate aminotransferase(AST) and nitric oxide(NO), liver index, thymus index and spleen index, and contents of tumor necrosis factor alpha (TNF-α ) and interleukin 1 beta(IL-113) in mice with immunological liver injury. The compatibility proportion of 18α- and 18β-glycyrrhizic acid was also screened. Results Compared with the model group, 18α- and 18β-glycyrrhizie acid remarkably lowered the activities of AST in the serum, and AST, ALT and NO in the liver homogenate, decreased TNF-α and IFN- contents in the liver homogenate, and improved liver index and thymus index. The differences of protective effect between the two glycyrrhizic acid groups were insignificant. The optimum compatibility proportion of 18α- and 18β-glycyrrhizic acid was 1:4, and the combination of 18α- and 18β-glycyrrhizic acid at the optimum compatibility proportion had better effect than 18α-glycyrrhizic acid or 18β-glycyrrhizic acid. Conclusion Both 18ot-glycyrrhizic acid and 18β-glycyrrhizic acid can protect mice from immunological liver injury, and they have similar effect. The optimum compatibility proportion of them is 1 : 4, and their combination in this proportion shows better effect on transaminases and NO levels in liver homogenate than 18α- and 18β-glycyrrhizic acid alone.
