摘要
目的对2014年5月由越南回国的女工所感染的麻疹病毒进行基因特征分析。方法采集病人的急性期咽拭子,通过RT-PCR和基因测序分析N基因碳末端450个核苷酸片段,利用BLASTn对序列进行相似性检索,分析麻疹病毒基因特征,并与参考株及沪191疫苗株进行比对,采用Neighbor-Joining方法构建麻疹核蛋白核苷酸种系进化树。结果获得1段相应样本N基因碳末端序列,上传至Gen Bank(KJ994346),该核苷酸序列与越南病毒株(Viet Nam:Ho Chi Minh City-AB928200)序列同源性100%,与WHO参考株(Manchester.UNK/30.94-AF280803)同一谱系,属于D8型,序列同源性为96%,氨基酸同源性为97%,其中产生5个基酸替换,第79位点发生变异,由异亮氨酸替代为苏氨酸,蛋白质极性发生改变,由疏水性变成亲水性。结论该病例为中山市首次发现的输入性D8型麻疹病毒感染病例。
Objective To analyze the genotype of measles virus isolated from a woman who returned to China from Vietnam. Methods Throat swab from the patient were collected during acute phase after infection and samples were assayed for preparing total viral RNA. About 450 nucleotides of the 3' terminus of nucleoprotein (N) gene was amplified with RT-PCR and sequenced. The sequence of the amplicons was analyzed and characterized with BLASTn, aligned with the WHO reference strain and Shanghai strain (NO.191). Phylogram tree analysis of this sequence was operated with Neighbor-Joining. Results Sequence of about 450 nucleotides of the 3' terminus of nucleoprotein (N) gene was obtained and uploaded to GenBank (KJ994346). Results of alignment showed that it formed a same clade withWHO reference strain (Manchester.UNK/30.94-AF280803), D8 genotype. Multi-sequence alignment analysis indicated that the nucleotide sequence shared 100% and 96% identity with Vietnam Strain (Viet Nam Ho Chi Minh City-AB928200) and WHO reference strain (Manchester.UNK/30.94-AF280803), respectively. The deduced amino acid sequence shared 97% identity with the WHO reference strainand 5 amino acids were substituted in the newly imported strain. The NO.79 amino acid, isoleucine, was substituted with threonine, which might lead to the polarity change of the protein from hydrophobic to hydrophilic. Conclusion The newly imported measles virus strain isgenotype D8, firstly found in Zhongshan.
