我国首例输入性D_9基因型麻疹病毒的分离和鉴定 被引量:51

D_9 Measles Virus was First Isolated from An Imported Measles Case in Sichuan Province of China

摘要 目的分离并鉴定我国首例输入性D9基因型麻疹病毒。方法使用Vero/SLAM细胞(非洲绿猴肾细胞/淋巴信号激活因子转染的非洲绿猴肾细胞)分离病毒,使用逆转录-聚合酶链反应扩增出编码核蛋白羧基末端450个核苷酸片段,通过对扩增产物进行核苷酸序列测定和分析,构建基因亲缘性关系树,进行遗传距离分析。结果四川省D9基因型麻疹病毒分离株MVi/Sichuan.CHN/07.09/1和世界卫生组织D9基因型代表株Victoria.AUS(维多利亚?澳大利亚)/12.99在基因亲缘性关系树上同属一个分支,核苷酸同源性为96.9%;和2008年流行于泰国和荷兰的D9基因型麻疹野病毒的核苷酸和氨基酸同源性分别为99.8%~100%和99.3%~100%;和中国大陆目前所使用的麻疹疫苗株沪191相比对,其核苷酸和氨基酸同源性分别为92.3%和90.7%;和中国目前流行的麻疹病毒绝对优势本土基因型H1基因型代表株相比对,其核苷酸和氨基酸同源性分别为90.8%和92.1%。结论输入中国四川省的麻疹病毒株为D9基因型。 Objective To isolate and identify the genotype and molecular characteristic of an imported D9 measles virus. Method To isolate the virus with Vero/SLAM cell line. And RT-PCR (reverse transcrip-tion-polymerase chain reaction) were performed to amplify the 450 nucleotide acids of carboxyl terminal of N (nucleoprotein,N) protein. The phylogenetic tree was constructed and the homology similarity was analyzed. Results Sichuan isolate MVi/Sichuan.CHN/07.09/1 was clustered within the same genotype group with WHO D9 genotype reference strain,the homology of nucleotide acid between Sichuan isolate and WHO D9 genotype reference strain was 96.9%. The homology of nucleotide acid and amino acid between Sichuan isolate and 2008 D9 genotype representative strain were 99.8%-100% and 99.3%-100% respectively. Compared with the Chinese measles vaccine strain,the homology of nucleotide acid and amino acid of Shanghai-191 were 92.3% and 90.7% respectively. Compared with the endemic H1 genotype representati.ve measles strain,the homology of nucleotide acid and amino acid were 90.8% and 92.1% respectively. Conclusion The imported virus was D9 genotype measles virus.
