摘要
目的天然药物以毒性低、安全范围广、药源丰富越来越受到人们的关注,本课题旨在研究姜黄素对甲状腺癌SW579细胞增殖、细胞周期和凋亡的影响,并探讨姜黄素抗癌的作用机制。方法甲状腺癌SW579细胞株经0~40μmol/L的不同浓度姜黄素作用12 h、24 h、48 h后,采用MTT法检测姜黄素对细胞增殖的抑制作用;经0~20μmol/L的不同浓度姜黄素作用7 d后,平板克隆形成实验检测姜黄素对甲状腺癌SW579细胞增殖的抑制作用;经0~20μmol/L的不同浓度姜黄素作用24 h后,采用流式细胞仪检测甲状腺癌SW579细胞周期;经0~20μmol/L的不同浓度姜黄素作用48 h后,采用流式细胞仪检测甲状腺癌SW579细胞凋亡率,并采用蛋白质印迹法检测甲状腺癌SW579细胞株中Cyclin B1、p21、bad和Bcl-xl的蛋白水平。结果 MTT结果显示姜黄素能显著抑制人甲状腺癌SW579细胞的生长增殖,抑制作用呈现时间及浓度依赖性;平板克隆实验提示,姜黄素作用甲状腺癌细胞后克隆形成数目下降,形成集落变小,并且呈浓度依赖性;流式细胞仪分析显示姜黄素能阻滞细胞周期于G0/G1期,并且呈浓度依赖性;流式细胞仪分析显示姜黄素能增加甲状腺癌细胞凋亡率;western blot检测显示姜黄素可抑制细胞周期蛋白Cyclin B1的表达,同时下调凋亡相关蛋白Bcl-xl,而姜黄素对P21和Bad的表达无影响。结论姜黄素能下调Cyclin B1、Bcl-xl的表达,显著抑制人甲状腺癌SW579细胞在体外的增殖。
Objective Natural drugs get more and more attentions because of their low toxicity and rich source, the aim of this study is to investigate the effect of cureumin on the proliferation, cell cycle distribution and apoptosis of thyroid carcinoma cells(SW579). Methods Thyroid carcinoma cells SW579 were cultured and treated with different content of cur- cumin (0 -40 μmol/L), the anti-proliferative activities at different time points were determined. MTY assays was used to test the anti-proliferative activities at 12,24 and 48 h in SW579 cells treated with(0 -40 μmol/L) curcumin;and other SW579 cells were treated with(0 -20 μmol/L) curcumin ,the cell cycle changes were detected by flow cytometic analysis 24 h later,the apoptosis ratio were detected by flow cytometic analysis and the levels of apoptosis related proteins Cy- clinB1 ,p21 ,Bad and Bcl-xl were analyzed by western blot 48h later,the anti-proliferative activities were determined by the colony formation assays 7 days later. Results The MTr assay revealed that curcumin had induced a dose-and a time- dependent decrease in cell viability. Colony counting indicated that curcumin had induced a dose-dependent decrease in the colony formation ability in SW579 cells. The flow cytometic analysis showed that SW579 cells treated with curcumin were arrested at the GO/G1 phase and the apoptosis ratio increased with a dose-dependent. Furthermore,western blot dem- onstrated that the curcumin decreased the expression of CyclinB1 and Bcl-xl in SW579 ceils, and had no influence on ex- pression of P21 and Bad. Conclusion The curcumin can significantly inhibit the proliferation of SW579 cells by down- regulating the expression of Cyclin B1 and Bcl-xl.
