摘要
目的 探讨Smad2、Smad3、Smad4在皮肤黑素瘤细胞中的表达情况。方法 应用反转录-实时荧光定量PCR方法和蛋白印迹方法,分别检测人皮肤黑素瘤A375细胞和人正常黑素细胞中Smad2、Smad3、Smad4的mRNA及蛋白表达。结果 A375细胞中Smad2、Smad3、Smad4的mRNA和蛋白表达均显著低于人正常黑素细胞(t=2.361-4.214,P〈0.01)。结论在黑素瘤细胞的转化生长因子β(TGF-β)/Smad信号通路中Smad2、Smad3、Smad4均出现表达下调,可能参与了皮肤黑素瘤发病。
Objective To investigate the expressions of Smad2, Smad3 and Smad4 in cutaneous melanoma. Methods Reverse transcription-real time polymerase chain reaction (RT PCR) and Western blotting were utilized to detect the expressions of A375 cell line in cutaneous melanoma and Smad2, Smad3 and Smad4 in normal melanocytes. Results The expressions of Smad2, Smad3 and Smad4 mRNA of A375 cells line were significantly lower than human normal m elanocytes (t = 2.361- 4.214, P〈0.01). Conclusion In TGF-β/Smad signaling pathways, the down regulation of Smad2, Smad3 and Smad4 may involve in the pathogenesis of cutaneous melanoma.
