摘要
目的研究转化生长因子-β1(transforming growth factor-β1,TGF-β1)、转化生长因子-β1Ⅱ型受体(transforming growthfactor-β1receptorⅡ,TGF-βRⅡ)和Smad4蛋白在原发性翼状胬肉组织中的表达及其意义。方法应用免疫组织化学SP法检测56眼翼状胬肉头部组织及8眼正常结膜组织中TGF-β1、TGF-βRⅡ及Smad4蛋白的表达,阳性表达率用阳性细胞占细胞总数百分率的平均值表示,利用统计学软件SPSS13.0分析本研究结果。结果TGF-β1在胬肉头部侵入角膜缘≤1mm、2-3mm和≥4mm翼状胬肉组和正常结膜组的阳性表达率利用分别为(21.08±3.63)%、(20.12±2.68)%、(21.55±3.07)%、(12.36±1.97)%;TGF-βRⅡ的表达率分别为(15.35±9.37)%、(21.07±10.96)%、(15.48±8.80)%、(33.77±9.31)%,Smad4蛋白的表达率分别为(14.38±4.02)%、(13.05±3.50)%、(13.13±4.10)%、(38.69±5.70)%,TGF-β1在各翼状胬肉组的表达率均高于正常结膜组(均为P〈0.001),TGF-βRⅡ、Smad4蛋白在各翼状胬肉组的表达率均低于正常结膜组(均为P〈0.001)。同一指标在翼状胬肉组间比较差异均无统计学意义(均为P〉0.05)。结论与正常结膜相比较,翼状胬肉组织中TGF-β1表达增加,TGF-βRⅡ及Smad4蛋白的表达降低,提示细胞生长调控在翼状胬肉的发生、发展中起着重要作用。
Objective To investigate the expression of transforming growth factor-beta one(TGF-β1),TGF-β1 receptor type Ⅱ(TGF-βRⅡ)and protein Smad4 in tissue of primary pterygium,and to further analyze their significance.Methods The expression of TGF-β1,TGF-βRⅡ and protein Smad4 were detected by immunohistochemical streptavidin-peroxidase(SP)technique in 56 pterygial head tissues and 8 normal conjunctival tissues.The positive expressive rate were expressed by average percentage-positive cells accounted for the total number of cells,and analyzed by statistical software SPSS version 13.0.Results The positive expressive rate of TGF-β1 in pterygium groups whose head invading to the cornea was less than or equal to 1 mm,between 2 mm and 3 mm,more than or equal to 3 mm,and in normal conjunctiva group were(21.08±3.63)%,(20.12±2.68)%,(21.55±3.07)% and(12.36±1.97)% respectively,that of TGF-βRⅡ was(15.35±9.37)%,(21.07±10.96)%,(15.48±8.80)% and(33.77±9.31)% respectively,and that of protein Smad4 was(14.38±4.02)%,(13.05±3.50)%,(13.13±4.10)% and(38.69±5.70)% respectively.The expression of TGF-β1 in every pterygium group was higher than that in normal conjunctiva group,there was significant difference(All P0.001);While the expression of TGF-βRⅡ and protein Smad4 in every pterygium group were lower than that in normal conjunctiva group,there was significant difference(All P0.001).And there was no statistical difference between every two pterygium groups with the same index(All P0.05).Conclusion Compared with normal conjunctiva,the expression of TGF-β1 increases in pterygium,and that of TGF-βRⅡ and Smad4 decrease in pterygium,which may play an important role in the occurrence and development of pterygium by regulating the cell growth.
