摘要
目的:探讨不同浓度TGF-β1抗体对翼状胬肉上皮细胞增殖的抑制作用。方法:通过组织块培养法进行人翼状胬肉上皮细胞的体外培养,以不同浓度TGF-β1抗体(0、6.25、12.5、25.0、50.0、100μg/ml)作用于传代培养的翼状胬肉上皮细胞,MTT法检测不同时期(24h、48h、72h)细胞增殖抑制情况;免疫组织化学SABC法染色检测增殖细胞核抗原(PCNA)的表达。结果:组织块培养法成功进行人翼状胬肉上皮细胞的原代和传代培养,并经角蛋白染色鉴定为上皮细胞;不同浓度TGF-β1抗体可明显抑制翼状胬肉上皮细胞的生长,且随浓度增加、时间延长,抑制率明显增高,PCNA的表达不同程度下降,各组间比较有统计学意义。结论:不同浓度TGF-β1抗体可明显抑制翼状胬肉上皮细胞生长,可作为治疗翼状胬肉的新型药物的研究方向。
Objective :To invesrigate the effect of TGF-β1 antibody on conjunctival epithelium cells and to explore a new way to prevent postoperative of pterygium .Methods:Human pterygium epithelium cells was cultured in vitro by tissue inoculation .Different concentration(0、6 .25、12 .5、25 .0、50 .0 、100μg/ml) of TGF-β1 antibody was used acting on the cells .Inhibitory effect of TGF-β1 antibody on pterygium epithelium cells was measured by MTT after 24 ,48 and 72 hours ,respectively .The antigen of PCNA was observed by SABC method of immunohistochem-istry .Results :Pterygium epithelium cells were cultured successfully by tissue inoculation ,and cells were identified immunoeytoehemieally with CK1 .Pterygium epithelium cells was inhibited by TGF-β1 antibody at different doses and the effect was more prominent as time went on .The expression of PCNA depressed as TGF-β1 antibody acting , and significant difference was found among groups .Conclusion :TGF-β1 antibody can inhibite the proliferation of hu-man pterygium epithelium cells ,and it offers a new research direction for the medicine on pterygium .
