摘要 目的:研究肿瘤坏死因子(TNF)-α对角质形成细胞糖皮质激素受体(GR)表达的影响。方法:采用反转录实时定量PCR和Western blotting观察经TNF-α作用后,培养的人角质形成细胞株HaCaT细胞的GRα、GRβ mRNA和蛋白质的表达情况。结果:TNF-α对HaCaT细胞GRα的表达无明显影响,但可上调HaCaT细胞GRβ的表达,并且呈剂量和时间依赖。结论:TNF-α可增加GRβ的表达,可能是炎性细胞因子参与诱发糖皮质激素皮肤耐受现象的内在机制之一。 Objective: To investigate the effects of TNF-α on the expression of glucocorticoid receptor (GRα and GRβ) in human keratinocytes. Methods: Cultured HaCaT cells were treated by TNF-α. The expressions of GRα and GRβ mRNA and protein were detected by reverse transcription fluorescence real time quantification PCR and Western blotting. Results:TNF-α had no effects on the expression of GRα, but up-regulsted the expression of GRB in HaCaT cells in a time-and dose-dependent manner. Conclusion: TNF-α could increase the expression of glucocorticiod receptor-β, which may be one of the mechanisms of glucocorticoid resistance induced by inflammatory cytokines.
