PARP-1与HMGA2在三阴型老年局部晚期乳腺癌中的表达及对预后的影响 被引量:5

摘要 目的通过免疫组织化学法实验观察PARP-1和HMGA2在老年局部晚期三阴型乳腺癌中的表达,探讨其在乳腺癌中表达的临床特点及其与预后的相关性。方法采用S-P免疫组织化学法检测80例三阴型老年局部晚期乳腺癌肿瘤标本和80例正常乳腺组织中PARP-1与HMGA2蛋白的表达情况。分析PARP-1和HMGA2在乳腺癌与正常乳腺组织中的表达差异;分析乳腺癌组织中PARP-1与HMGA2表达的相关性及其分别与原发肿瘤直径、腋窝淋巴结转移及病理组织学分级的相关性。并且通过Kaplan-Meier生存曲线法比较两者阳性组和阴性组患者的预后情况。结果在乳腺癌中PARP-1阳性表达率为84%显著高于正常乳腺组织的21%;HMGA2阳性表达率为61%高于正常乳腺组织的26.3%,PARP-1与HMGA2在老年局部晚期三阴型乳腺癌中的阳性表达与腋窝淋巴结转移情况及5年无瘤生存时间具有相关性(P<0.05)。PARP-1与HMGA2在各分子亚型乳腺癌组织中的表达具有相关性(P<0.05)。结论 PARP-1与HMGA2在老年局部晚期乳腺癌的浸润、转移中存在相互作用,是反映乳腺癌进展的重要生物学指标。PARP-1与HMGA2高表达的患者易发生淋巴结转移,并且无瘤生存时间缩短,预后较差。 Objective Through the immunohistochemical method experiment,this paper aims to observe the expressions of PARP-1and HMGA2 in senile patients with triple-negative locally advanced breast cancer and to discuss their clinical significance.Methods With the S-P immunohistochemical method,it detected the PARP-1and HMGA2 protein expressions in both 80 cases of breast cancer postoperative tumor specimens and another 80 cases of normal breast tissue with a distance of at least 5cm from the tumor.We analyzed the expression difference of PARP-1and HMGA2 in breast cancer and normal breast tissue.Spearman rank test was used to analyze the correlation of their expressions,and the correlation with primary tumor diameter,auxiliary lymph node metastasis and pathological classification,respectively.We also made a comparison of the prognosis between the positive and negative patient groups through the Kaplan-Meier survival curve method.Results It showed that the positive expression rate of PARP-1in breast cancer(84%)was significantly higher than that in paracancerous normal tissues(21%),while the positive expression rate of PARP-1in breast cancer(61%)was higher than that in paracancerous normal tissues(26%).Two of them had remarkable relation to the lymph node metastasis and disease-free survival time of 5years(P〈0.05).Besides,the expression of PARP-1and that of HMGA2 were obviously related(P〈0.05).Conclusion The expressions of PARP-1and HMGA2 play cooperative roles in the infiltration and metastasis of breast cancer,which also serve as important biological parameters for judging the breast cancer progression.Patients with up-regulated expression of PARP-1and HMGA2 are prone to have lymph node metastasis and poor prognosis after operation.
