不同分子分型乳腺癌组织中HSF1的表达情况及其与预后的相关性 被引量:1

Analysis of HSF1 Expression and Its Correlation with Prognosis in Different Molecular Type Breast Cancer Tissues

摘要 目的探讨不同分子分型乳腺癌组织内热休克转录因子1(HSF1)的表达情况及其与预后的关系。方法回顾性分析186例女性乳腺癌患者的病历资料,依照雌激素受体(ER)、孕激素受体(PR)以及人表皮生长因子受体2(HER2)表达水平的不同,把所有患者划分成Luminal A型组(n=68)、Luminal B型组(n=46)、HER2型组(n=34)与三阴性组(n=38)。采用免疫组织化学染色法检测各组患者乳腺癌组织内的HSF1、热休克蛋白70(HSP70)、抗凋亡蛋白B细胞淋巴瘤因子-2(Bcl-2)、巨噬细胞移动抑制因子(MIF)表达水平,以Pearson相关性分析法分析HSF1与HSP70、Bcl-2、MIF的关系;同时随访3年,运用Kaplan-Meier法分析HSF1阳性、阴性表达患者的生存率。结果Luminal A型组与Luminal B型组患者乳腺癌组织内HSF1、HSP70、Bcl-2与MIF阳性表达率均低于HER2型与三阴性组患者,差异有统计学意义(P<0.05);乳腺癌组织内HSF1与HSP70、Bcl-2与MIF表达呈正相关(γ=0.358、0.405、0.426,P<0.05)。HSF1阳性表达患者的3年生存率[44.25%(50/113)]低于阴性表达患者[67.12%(49/73)],差异有统计学意义(P<0.05)。结论HSF1在HER2型与三阴性乳腺癌中的表达水平较高,且HSF1阳性表达患者的3年生存率更低,预后更差。 Objective To investigate the expression of heat shock transcription factor 1(HSF1)in different molecular types of breast cancer and its relationship with prognosis.Methods Clinical data of 186 female breast cancer patients were retrospectively analyzed.According to the expression levels of estrogen receptor(ER),progesterone receptor(PR)and human epidermal growth factor receptor 2(HER2),all patients were divided into luminal type A group(n=68),luminal type B group(n=46),HER2 type group(n=34)and triple negative group(n=38).The expression levels of HSF1,heat shock protein 70(HSP70),anti apoptotic protein B-cell lymphoma factor-2(Bcl-2)and macrophage migration inhibitory factor(MIF)in breast cancer tissues of patients in each group were detected by immunohistochemical staining,and the relationship between HSF1,HSP70,Bcl-2 and MIF was analyzed by Pearson correlation analysis;At the same time,the survival rate of HSF1 positive and negative patients was analyzed by Kaplan Meier method after 3 years of follow-up.Results The positive expression rates of HSF1,HSP70,Bcl-2 and MIF in breast cancer tissues of patients with luminal type A and luminal type B were lower than those of patients with HER2 and triple negative(P<0.05);The expression of HSF1,HSP70,Bcl-2 and MIF in breast cancer was positively correlated(γ=0.358,0.405,0.426,P<0.05).The 3-year survival rate[44.25%(50/113)]of patients with positive HSF1 expression was lower than that of patients with negative HSF1 expression[67.12%(49/73)],the difference was statistically significant(P<0.05).Conclusion HSF1 is highly expressed in HER2 and triple negative breast cancer,and the 3-year survival rate and prognosis of patients with HSF1 positive expression are lower.
