摘要
目的以环磷酰胺所致免疫功能低下小鼠为模型,探讨不同频率电针(EA)刺激对其T细胞亚群和β-内啡肽的影响。方法应用流式细胞术和放射免疫方法测定在不同频率电针刺激下小鼠T细胞亚群、β-内啡肽(β-EP)含量的变化。结果连续电针3d(每天1次)后,2Hz、100Hz和2/100 Hz EA对外周血中CD+4及CD+8均无明显影响,而2/100 Hz EA可明显升高CD+4/CD+8(P<0.05),但2Hz和100Hz EA则没有;与环磷酰胺组相比,2 Hz EA和100 Hz EA的下丘脑、延髓和脾脏的β-EP均变化不明显,2/100Hz EA在下丘脑及延髓的β-EP均变化不明显,但脾脏的β-EP显著升高(P<0.01),而且与2 Hz EA和100 Hz EA的β-EP相比均明显升高(P<0.05,P<0.01)。结论电针对小鼠T细胞亚群和β-内啡肽(β-EP)含量的作用均存在频率特异性,这将为临床应用电针治疗与免疫相关疾病提供理论和实验依据。
Objective To explore the effect of electroacupuncture(EA) with different frequencies on T cell subsets and β-endorphin(-EP) in cyclophosphamide(CP) mice. Methods Application of flow cytometry techniques and RIA were used to detect the effect of EA at different frequency,to confirm the effect of EA on the level of splenic T cell subsets and-EP in CP mice.Results After three days of successive EA with frequencies of 2Hz,100 Hz and 2/100 Hz,blood of CD4+and CD8+did not significantly change in 2Hz,100 Hz and 2/100 Hz; while CD4+/CD8+was significantly increased when treated with 2/100 Hz EA(P〈0. 05),but not in 2Hz and 100 Hz. Compared with the CP group,-EP did not significantly change in hypothalamus,medulla oblongata and spleen in 2 Hz EA and 100 Hz EA,as to 2/100 Hz EA-EP did not significantly change in hypothalamus,medulla oblongata,but significantly changed in the spleen(P〈0. 01),and the level of-EP were significantly higher than 2 Hz EA and 100 Hz EA(P〈0. 05,P〈0. 01). Conclusion EA has the effect of T cell subsets and-EP,and the existence of frequency responsiveness. This will provide the theoretical and practical basis for clinical application of EA.
