大鼠左侧颈总动脉与颈外静脉吻合分流性肺动脉高压模型建立 被引量:1

Establishment model of new pattern and arteriovenous shunt of pulmonary artery hypertension in rats

摘要 目的建立大鼠左颈总动脉与左侧颈外静脉吻合分流性肺动脉高压模型。方法成年雄性Wistar大鼠随机分为三组:吻合组(A组,n=15),结扎组(B组,n=15),正常组(C组,n=15)。饲养3月后测定血流动力学指标,右心室/左心室+室间隔质量比,肺组织切片病理变化,ELISA方法测定血浆ET-1浓度,荧光定量PCR测定肺组织表达内皮素-1mRNA的变化。结果与B,C两组相比,A组:右心室收缩压力明显升高,血清内皮素-1水平升高,荧光定量PCR结果显示A组内皮素-1mRNA的相对表达量是B组的2.601倍,是C组的3.373倍,B组是C组的1.297(P<0.05)。结论通过分离左颈总动脉及左颈外静脉切断后,近心端吻合,可成功建立模拟先天性心脏病的分流性肺动脉高压模型,作为肺动脉高压标志的内皮素-1在此模型中处于高表达状态。 Objective To establish arteriovenous shunt of pulmonary artery hypertension by anastomosis of the left common carotid artery and left external j ugular vein.Methods Adult male Wistar rats were randomly divided into three groups:anastomosis group (group A,n=15),ligation group (group B,n=15),normal group (group C,n=15).After feeding 3 months,hemodynamic index,pathological changes of myocardial tissue and lung biopsy,the plasma ET-1 con-centration,and the change of lung tissue endothelin-1 mRNA were determined.Results Compared with group B and C, the right ventricular systolic pressure increased significantly in group A.The concentration of plasma endothelin 1 was higher significantly in group A than group B and C (P〈0.05).Fluorescence quantitative PCR results showed that the relative expression level of endothelin 1 mRNA in group A is 2.601 times of that in group B,3.373 times of that in group C,and that in group B is 1.297 times of that in the group C (P〈0.05).Conclusion The arteriovenous shunt of pulmonary artery hypertension can be established successfully through anastomosis of the left common carotid artery and left external j ugular vein.Endothelin-1 is in a state of high expression in this model.
