结肠癌组织LSD1表达对结肠癌LoVo细胞转移潜能影响研究 被引量:5

Expression of LSD1 in colon carcinoma tissues and its effects on metastatic potential of LoVo cells

摘要 目的:研究赖氨酸特异性组蛋白去甲基化酶1(lysine specific demethylase 1,LSD1)在人结肠癌组织中的表达,及其对人结肠癌LoVo细胞株转移潜能的影响。方法:收集2007-06-01-2011-08-31南通大学附属医院52例结肠癌石蜡组织标本,免疫组化方法检测52例组织中LSD1的表达情况并分析其临床意义;瞬时转染LSD1特异性短发卡RNA(shRNA)干扰质粒下调LSD1的表达,通过细胞划痕实验、Transwell迁移实验和Transwell侵袭实验观察下调LSD1的表达后LoVo细胞迁移及侵袭行为的变化。结果:LSD1在结肠癌组织中阳性表达率为67.3%(35/52),在癌旁组织中为37.5%(9/24),χ2=5.958,P=0.014。结合临床资料分析显示,LSD1的表达与分化程度(P=0.001)、淋巴结转移(P=0.011)及Duke分期(P=0.024)有关,而与患者性别、年龄、肿瘤部位、肿瘤直径、大体类型、浸润深度及有无远处转移无关。细胞划痕结果显示,转染组细胞迁移能力明显受抑制;Transwell迁移实验结果显示,Control组和转染LSD1-shRNA组穿过膜细胞数分别为220.25±6.75和40.875±1.813,转染LSD1-shRNA组穿过膜细胞数明显减少,z=3.787,P<0.001;Transwell侵袭实验结果显示,Control组和转染LSD1-shRNA组穿过膜细胞数分别为222.75±6.25和33.625±1.75,转染LSD1-shRNA组穿过膜细胞数明显减少,z=3.782,P<0.001。结论:LSD1表达与结肠癌的恶性程度及转移潜能密切相关,下调LSD1表达能抑制结肠癌LoVo细胞的转移潜能。 OBJECTIVE:To study expression of lysine specific histone demethylase I(LSD1) in colon carcinoma tissues and its effects on metastatic potential of LoVo cells. METHODS: Totally 52 paraffin section samples of colon carcinoma tissues (obtained from Affiliated Hospital of Nantong University, 2007-06-01--2011-08-31)were detected for the expression of LSD1 and its clinical significance; expression of LSD1 was down regulated by transient transfeetion of LSDl-specific short hairpin RNA(shRNA) interference plasmid, changes of migration and invasion of LoVo cells were ob- served by cell scratch test and transwell migration experiment as well as Transwell invasion experiment after LSD1 activi ty was inhibited. RESULTS: LSD1 was highly expressed in colon carcinoma tissues(67.3%), but it was low in adjacent tissues (37.5 %, X2 = 5. 958, P= 0. 014). Combined with analysis of clinical data it showed that the differences of expression of LSD1 were statistically significant in colon carcinoma at various degrees of differentiation (P = 0. 001), with or without lymph node metastasis (P=0. 011) and at different Duke stages (P=0. 024) ,while the expression of LSD1 was unrelated to patients' gender, age, tumors' site, diameter, type, depth of invasion and distance of metastasis. Results of cell scratch test showed that migration ability of cells in transfeetion group was inhibited obviously. Results of transwellmigration experiment showed that numbers of cells passing through the membrane were 220. 25± 6. 75 in the control group and 40. 875 ±1. 813 in the LSDI-shRNA transfection one,and cell number of the transfection one was significantly reduced (z=3. 787, P〈0. 001). Results of transwell invasion experiment showed that numbers of cells passing through the membrane were 222.75 ± 6.25 in the control group and 33. 625 ±1.75 in LSDI-shRNA transfection group, and cell number of the transfeetion one was significantly reduced (z = 3. 782, P〈 0. 001).
