摘要
目的:探讨人前列腺癌细胞转移潜能与其环氧化酶-2(COX-2)表达之间的关系,及环氧化酶-2与前列腺癌患者预后的关系。方法:用运免疫组化及反转录聚合酶链式反应(RT-PCR)检测两种不同转移潜能人前列腺癌细胞PC3及PC3M中COX-2mRNA及其蛋白的表达以及在肿瘤坏死因子-α(TNF-α)刺激下两者的表达是否有所变化。结果:PC3细胞在无TNF-α刺激时,检测不到COX-2mRNA的表达,其蛋白表达也为阴性,而在TNF-α刺激4h后,COX-2mRNA及其蛋白的表达均为阳性。而在PC3M细胞中无TNF-α刺激时,COX-2mRNA及其蛋白的表达已为阳性,在TNF-α刺激4h后,COX-2mRNA及其蛋白的表达均有增强。结论:COX-2可能在前列腺癌的发生发展中起着重要的作用,抑制COX-2的表达,或许能够降低前列腺癌的恶性程度甚至能够阻止其发生。这对前列腺癌患者生存期内创造良好的生活质量及前列腺癌的防治提供有力的理论依据。
AIM:To investigate the relationship between human prostate cancer cells with d ifferent metastatic potential and expression of COX-2. METHODS:Expression of cyclooxygenase-2 and it's regulation by tumor necrosis factor-αin PC3 and PC3M were detected by reverse transcription-PCR and immun ohistochemistry. RESULTS:We observed that the COX-2 mRNA level and COX-2 protein were undetec table in PC3 cells but it was positive at 4 h after stimulation with TNF-α.In PC3M, there were a gradual increase in COX-2 expression up to 4 h after stimula tion with TNF-αcompared with the beginning.
