摘要
目的探究套细胞淋巴瘤C-myc基因、LSD1表达及临床意义。方法回顾性分析2010年1月~2015年12月就诊于我院的30例套细胞淋巴瘤(MCL)患者的临床资料,将其纳入研究组,并将同时期的30例增生性淋巴结炎患者临床资料纳入对照组。采用SP免疫组织化学方法检测两组病例病理组织中C-myc、LSD1蛋白表达情况,并应用小干扰RNA(siRNA)沉默套细胞淋巴瘤Jeko-1细胞的C-myc基因,Western blot检测C-mycsiRNA作用后凋亡相关蛋白C-myc、Bcl-2、Procaspase3表达水平。分析C-myc、LSD1蛋白表达与套细胞淋巴瘤临床病理分期的相关性及C-mycsiRNA对套细胞淋巴瘤Jeko-1细胞凋亡相关蛋白C-myc、Bcl-2、Procaspase3的影响。结果经Spearman相关性分析显示,C-myc、LSD1蛋白表达与MCL临床病理分期无相关性(P>0.05)。C-mycsiRNA处理Jeko-1细胞24 h后,Western blot检测显示凋亡相关蛋白C-myc、Bcl-2、Procaspase3表达呈浓度依赖性下降。结论C-myc、LSD1蛋白表达与MCL临床病理分期无相关性,干扰Jeko-1细胞C-myc基因后可下调凋亡相关蛋白C-myc、Bcl-2、Procaspase3表达水平,C-myc基因可能成为靶向治疗的新靶点。
Objective To explore the expression of C-myc gene and LSD1 in mantle cell lymphoma and its clinical significance.Methods The clinical data of 30 patients with mantle cell lymphoma(MCL)who were treated in our hospital from January 2010 to December 2015 were retrospectively analyzed and included as the study group,and 30 patients with proliferative lymphadenitis in the same period were included as the control group.SP immunohistochemistry was used to detect the expression of C-myc and lysine specific demethylase 1(LSD1)proteins in the pathological tissues,and small interfering RNA(siRNA)was used to silence the C-myc gene of MCL Jeko-1 cells.Western blot was used to detect the expression of apoptosis-related proteins C-myc,Bcl-2,and Procaspase3 after C-myc siRNA treatment.The correlation between the expression of C-myc and LSD1 proteins and the clinicopathological stage of MCL and the effect of C-myc siRNA on the apoptosis-related proteins C-myc,Bcl-2,and Procaspase3 of MCL Jeko-1 cells was analyzed.Results Spearman correlation analysis showed that there was no correlation between C-myc and LSD1 protein expression and clinical pathological stage of MCL(P>0.05).After C-myc siRNA treatment of Jeko-1 cells for 24 hours,Western blot showed that the expression of apoptosis-related proteins C-myc,Bcl-2,and Procaspase3 decreased in a concentration-dependent manner.Conclusion There is no correlation between the expression of C-myc and LSD1 proteins and the clinical pathological stage of MCL.Interfering with the C-myc gene in Jeko-1 cells can down-regulate the expression of apoptosis-related proteins C-myc,Bcl-2,and Procaspase3.The C-myc gene may become a new target for targeted therapy.
