红毛五加多糖对体外人胃癌细胞的抑制作用特点及机制 被引量:5

摘要 目的 研究红毛五加多糖对胃癌细胞的抑制作用特点 ,并探讨该多糖的抑癌机制。方法 选取MTT法检测该多糖对癌细胞和正常人细胞的抑制作用特点 ,采用流式细胞仪以检测基因蛋白表达。结果 该多糖选择性地抑制胃癌细胞的活性和功能状态 ,而对正常人外周血淋巴细胞和人胚腱细胞的活性和功能状态无影响。对癌基因 c- myc蛋白表达与空白对照组比较没有显著性差异。结论 红毛五加多糖可能不是通过抑制癌基因 c- myc蛋白表达而达到抑制胃癌细胞增殖效应。由于该多糖抑制胃癌细胞的同时对健康人细胞无影响 。 Objective To investigate the inhibitory character and the mechanism of Acanthopanax giraldii Harms Var Hispidus Hoo polysaccharides (AGP) on the in vitro human gastric cancer cell line.Methods MTT approach that shows the character of the inhibitory effect and flow cytometre which can explore the mechanism were adopted.Results The polysaccharides selectively inhibited the activity and function of the gastric cancer cell,while that of the lymphocyte of peripheral blood in healthy people and human embryonic tenocyte was not affected.The protein expression of the c myc oncogene has no significant differences between the blank group and the AGPI group.Conclusion The inhibitory effect of the polysaccharides may not be induced by the protein change of c myc oncogene.These polysaccharides could be safely utilized to prevent and cure cancer.
