壬基酚对仔鼠脑皮质增殖细胞核抗原表达的影响 被引量:2

摘要 目的母鼠孕期和哺乳期暴露NP,观察其对子代脑皮质增殖细胞核抗原(PCNA)的影响。方法试验分为4组:即对照组,NP25、50、100组。母鼠受孕第6天到出生后21 d染毒壬基酚,21、60 d分别剖杀检测PCNA的表达,并分析上述变化与NP暴露之间的关系。结果与对照组比较,低、中、高剂量组21、60 d PCNA阳性细胞数明显增多(F=476.34、1077.55,P<0.01),光密度增高(F=44.46、7.58,P<0.01)随着给药剂量和给药时间的增加,PCNA阳性细胞数目逐渐增多,21、60 d阳性细胞数与NP暴露剂量呈正相关(r=0.87、0.60,P<0.05)。结论 NP孕期暴露使仔鼠脑组织PCNA的表达增加,即处于增殖状态的细胞增多,提示NP对脑组织有损伤作用。 Objective To observe the possible effect of Nonylphenol on proliferating-cell nuclear antigen(PCNA) expression in the brain cortex of pups from dams exposed during gestational and lactational periods.Methods Pregnant dams were randomly assigned to 4 groups including NP treatment at dose levels of 25 mg/kg/day(low dose),50 mg/kg/day(middle dose),100 mg/kg/day(high dose) and groundnut oil treatment alone(vehicle control),respectively.The pregnant dams were housed individually and fed intragastrically.NP exposure time was limited from gestational day 6 to postnatal day 21.The expression of PCNA in the brain cortex of rat pups at PND 21 and PND 60 was detected using immunohistochemistry,and the association between NP exposure dose and these alterations in PCNA expression was analyzed as well.Results PCNA-positive cells(F =476.34、1077.55,P 〈 0.01) and integrated optical density(IOD)(F = 44.46、7.58,P 〈 0.01) of pup cerebral cortex at PND 21 and 60 in low-dose,middle-dose and high-dose NP treatment groups were higher than the control group(P 〈 0.05).NP exposure dose and time were associated with PCNA-positive cells in a dose-response manner.A positive and significant relationship between NP exposure dose and PCNA-positive cells was revealed(r = 0.873、0.606,P 〈 0.05).Conclusion Exposure to NP during gestation might induce an increase in PCNA expression in the brain of pup rats,namely an increase in cells in a state of proliferation,which indicated an adverse effect of NP on brain.
