摘要
用氧化型低密度脂蛋白(Ox-LDL)诱导人脐静脉平滑肌细胞(HUVSMC)增殖模型,探讨三硬脂酸甘油酯、三油酸甘油酯和三亚油酸甘油酯对Ox-LDL诱导的细胞的影响。采用MTT法测定HUVSMC的增殖,划线伤口愈合法测定HUVSMC迁移能力来观察不同甘油三酯对Ox-LDL诱导HUVSMC增殖与迁移的影响,并用Western Blotting法测定平滑肌细胞增殖细胞核抗原(PCNA)的表达。研究结果表明:对质量浓度为50 mg/L的Ox-LDL所诱导的HUVSMC的增殖、迁移,三硬脂酸甘油酯有明显促进作用并使平滑肌细胞PCNA的表达上调,而三油酸甘油酯和三亚油酸甘油酯有抑制作用并使平滑肌细胞PCNA的表达下调。
The effects of tristearin, triolein and trilinolein on the proliferation, migration and expression of proliferating cell nuclear antigen (PCNA) of human umbilical vein smooth muscle cells ( HUVSMC ) in- duced by oxidized low - density lipoprotein ( Ox - LDL) were investigated. Methyl thiazolyl tetrazolium (MTT) assay was used to determine the proliferation of HUVSMC, and Crossed wound healing assay was used to determine the migration ability of HUVSMC to observe the effects of different triglycerides on the proliferation and migration of HUVSMC induced by Ox - LDL, and Western blotting method was used to determine the PCNA expression of HUVSMC. The results showed that the tristearin acted a significant effect on promoting the proliferation and migration of HUVSMC induced by 50 mg/L of Ox - LDL and the PCNA expression, while triolein and trilinolein had inhibitory effects.
