摘要
目的探讨人乳头瘤病毒(Human papillomavirus,HPV)感染,Rb蛋白和Mcl-1蛋白表达的变化同非小细胞肺癌(Non-small-cell lung cancer,NSCLC)的相关性。方法分别应用PCR、免疫组化方法 (IHC)检测NSCLC组(79例)和肺良性病变组(11例)中HPV DNA表达和Rb蛋白、Mcl-1蛋白的表达。结果在NSCLC组织中,HPV DNA的PCR检测阳性检出率为46.8%,肺良性病变组阳性检出率为9.1%(P<0.05),HPV阳性表达与组织学类型、分化程度及吸烟相关。NSCLC中HPV阳性组Rb蛋白表达阳性率(37.8%)显著低于阴性组(61.9%)(P<0.05)。NSCLC中HPV阳性组和阴性组Mcl-1蛋白表达阳性率分别为59.5%和35.7%(P<0.05)。肺良性病变组中Rb蛋白表达阳性率为72.7%而Mcl-1蛋白未表达。结论 NSCLC的发生发展可能与HPV感染后通过抑制Rb蛋白通路同时促进Mcl-1蛋白通路表达有关。
Objective To study the relationship between HPV ( Human papillomavirus, HPV) infection, the expression of Rb&amp;Mcl-1 protein and the non-small cell lung cancer ( Non-small-cell Lung Cancer, NSCLC) . Methods PCR and Immunohistochemis-try (IHC) were used to detect HPV DNA expression and protein expression of Rb and Mcl-1 in NSCLC group (79 cases) and benign lung lesion group (11 cases). Results The positive rate of HPV in NSCLC group is 46.8% , and the positive rate of HPV in lung benign lesion group is 9.1% (P〈0.05). HPV positive expression is related to histological type, degree of differentiation and smoking. Rb protein positive expression in HPV-positive group ( 37.8%) is significantly lower than the negative group ( 61.9%) in NSCLC ( P〈0.05 ) . Mcl-1 protein positive expression in HPV-positive group ( 59.5%) is significantly lower than the negative group (35.7%) in NSCLC (P〈0.05). In benign lesion group, the rate of Rb protein expression is 72.7%, but Mcl-1 protein is not ex-pressed. Conclusion Development of NSCLC is related to HPV infection by inhibiting the Rb protein pathway and promoting Mcl-1 protein pathway.
