摘要
目的:探讨急性心肌缺血再灌注大鼠心功能的改变及心肌纤维化对其影响。方法:建立大鼠急性心肌缺血再灌注模型,利用心脏超声和血流动力学检测其心功能状态。用水解法检测心肌羟脯氨酸含量。分别用免疫印迹和酶联免疫吸附剂测定法检测心肌和血清中TNF-α的表达。结果:急性心肌缺血再灌注大鼠早期出现心脏舒张功能降低,在灌注第3天时尤为显著。心肌纤维化发生在缺血再灌注区,且该部位胶原容积分数显著增高,与心肌TNF-α表达规律相同。同时心肌羟脯氨酸含量和血清TNF-α在急性缺血再灌注时也显著增加。结论:急性心肌缺血再灌注大鼠心肌纤维化降低早期心脏舒张功能,而炎症因子TNF-α的异常增高是急性心肌缺血再灌注心肌纤维化的可能因素之一。
Objective:To discuss the cardiac function changes and the myocardial fibrosis effects in acute myocardial ischemia/reperfusion rats .Methods :Acute myocardial ischemia/reperfusion rats models were estab-lished for detecting their heart function by echocardiography and hemodynamic .Water solution method was used to detect expression of myocardial hydroxyproline .And the expression of TNF-αin the myocardial and serum was de-tected by western blot and enzyme-linked immunosorbent method .Results :Early heart diastolic function was lower in acute myocardial ischemia/reperfusion rat ,significantly in the third day after perfusion .Myocardial fibrosis oc-curred in ischemia/reperfusion areas ,and the collagen volume fraction was significantly increased in this area .It was identical to expression of TNF-αin myocardial .Meanwhile ,myocardial hydroxyproline content and TNF-αin serum also significantly increased in acute ischemia/reperfusion .Conclusion:Myocardial fibrosis results to deterioration of early heart diastolic functionin in rats with acute myocardial ischemia/reperfusion .And the expression of inflamma-tory cytokine TNF-αis one of the possible factors of myocardial fibrosis in acute myocardial ischemia/reperfusion .
