摘要 目的:探讨慢病毒介导肝癌缺失基因-1(Deleted in liver cancer-1,DLC-1)转染结肠癌SW480细胞系对β-catenin表达水平的影响及意义。方法:用携带DLC-1基因的重组慢病毒转染结肠癌SW480细胞系;采用Real-time PCR及Western blot法检测SW480细胞系中DLC-1及β-catenin的表达水平,MTT法检测SW480细胞的增殖活性,流式细胞仪检测细胞的凋亡率,Transwell小室法检测SW480细胞迁移和侵袭能力。结果:DLC-1重组慢病毒转染SW480细胞系中,过表达的DLC-1下调β-catenin表达水平,抑制SW480细胞的增殖、迁移及侵袭,并诱导其凋亡。结论:DLC-1基因可能通过下调β-catenin的表达来调节结肠癌SW480细胞系的增殖、凋亡、迁移和侵袭等生物学行为,DLC-1在结肠癌的发生、发展中可能发挥了十分重要的作用。 Objective:To investigate the biological roles of Deleted in liver cancer-1 ( DLC-1 ) gene in biological behaviors of human colon cancer cell line, and to explore the underlying molecular mechanisms of the actions. Methods: The recombinant Lentiviral vectors containing DLC-1 gene were constructed to transfect the human colon cancer cell line SW480. Real-time PCR and Western blot analysis were performed to evaluate the expression levels of DLC-1 and β-catenin in the transfected colon cancer cell line. In addition, cell proliferation assay, cell apoptosis analysis, cell migration assay and invasion assay were performed on the transfected SW480 cell line. Results: Both Real-time PCR and Western blot analysis revealed overexpression of DLC-I gene in the transfected SW480 cell line, and expression level of β-catenin in which was decreased. Proliferation, migration and invasion of SW480 cells were significantly inhibited and number of apoptosis cells was increased. Conclusion: DLC-1 gene overexpression may inhibit cell growth, cell migration as well as invasion and induce cell apoptosis in colon cancer via increasing expression level of β-catenin. DLC-1 probably play an im- portant role in progress and metastasis of colon cancer.
