摘要
目的:检测卵巢上皮性癌组织中黏着斑激酶(FAK)和肝癌缺失基因-1(DLC1)mRNA的表达。方法:采用逆转录聚合酶链反应检测12例正常卵巢和32例卵巢上皮性癌组织中FAK与DLC1 mRNA的表达。结果:卵巢上皮性癌组织中FAK mRNA的表达水平(0.81±0.33)高于正常卵巢组织(0.24±0.15),2者相比,差异有统计学意义(t=2.979,P=0.007);FAK mRNA的表达水平与卵巢上皮性癌的分化程度、淋巴结转移和腹水形成有关(t/F=3.405,2.223和2.330,P均<0.05)。卵巢上皮性癌组织中DLC1 mRNA的表达水平(0.30±0.11)低于正常卵巢组织(0.68±0.17),2者相比,差异有统计学意义(t=-3.284,P=0.013);DLC1 mRNA的表达与卵巢上皮性癌的临床分期、淋巴结转移和腹水形成有关(t/F=3.156,2.944和2.470,P均<0.05)。卵巢上皮性癌组织中FAK与DLC1 mRNA的表达有关联(rP=0.369,P=0.025)。结论:FAK的过表达和DLC1低表达或表达缺失可能与卵巢上皮性癌的发生、发展有关;联合检测2者有助于判断卵巢上皮性癌的恶性程度。
Aim: To detect the expression of FAK mRNA and DLCI mRNA in epithelial ovarian cancer tissue. Methods:RT-PCR was used to detect the expression of FAK mRNA and DLCI mRNA in epithelial ovarian cancer and normal o- varian tissue. Results: The expression level of FAK mRNA in epithelial ovarian cancer tissue (0.81 ±0. 33 )was signifi- cantly higher than that in normal ovarian tissue [ (0.24 ± 0.15 ) , t = 2. 979, P = 0. 007 ] ; the expression level of FAK mRNA was related with differentiation,lymphatic metastasis and ascites formation ( t/F = 3. 405,2. 223 and 2. 330, all P 〈 0.05 ). The expression level of DLC1 mRNA in epithelial ovarian cancer tissue( 0.30 + O. 11 ) was significantly lower than that in normal ovarian tissue [ (0.68 ±O. 17 ) , t = - 3. 284, P = 0. 01 3 ]. The expression level of DLC1 mRNA was related with clinical stage, ascites formation and lymphatic metastasis ( t/F = 3.156,2. 944 and 2. 470, all P 〈 O. 05 ). The expression of FAK mRNA had association with that of DLC1 mRNA ( re = O. 369, P = 0. 025 ). Conclusion : The overexpression of FAK and the underexpression or loss of DLC1 may play important roles in the generation and development of epithelial ovarian cancer;combined detection of FAK and DLC1 may be helpful in predicting the maligant degree of epithelial ovarian eaneer,
