摘要
目的探讨胃癌患者癌组织中肝癌缺失基因(DLC-1)的表达水平及其与患者的临床病理特征的关系。方法对本院收集的60例原发性胃癌患者的术后癌组织标本、癌旁组织及在本院进行胃镜取组织活检诊断正常的胃组织30例进行逆转录-聚合酶链反应检测,比较三种胃组织标本中DLC-1 m RNA的表达水平差异。检测DLC-1蛋白在胃癌组织中的表达及与胃癌患者的临床病理特征的关系。结果胃癌组织中的DLC-1 m RNA灰度值为0.22±0.08,显著低于癌旁组织(0.41±0.11)和胃正常组织(0.63±0.13),差异均有统计学意义(P<0.05)。60例胃癌患者中,DLC-1蛋白阳性表达24例(60%),DCL-1蛋白阳性表达率与胃癌患者的年龄、性别、病灶大小无关(P>0.05);低分化胃癌患者的DLC-1蛋白表达率为25%、TNM分期Ⅲ+Ⅳ期患者的DLC-1蛋白表达率为29.17%、浸润深度T3+T4的患者的DLC-1蛋白表达率为33.33%、发生淋巴结转移的患者的DLC-1蛋白表达率为16.67%、胃癌发生远处转移的患者的DLC-1蛋白表达率为20.83%均显著的低于相对应的高+中分化患者、TNMⅠ+Ⅱ期患者、浸润深度T1+T2、无淋巴结转移的患者、无远处转移的患者(P<0.05)。结论胃癌患者癌组织中DLC-1的表达水平较癌旁组织、正常胃组织显著降低,DLC-1蛋白与低分化、肿瘤分期、浸润深度、淋巴结转移、远处转移具有相关性。
Objective Discussion of deleted in liver cancer tissue of patients with gastric cancer gene(DLC-1) expression levels and its relationship with clinical pathological features of patients. Methods Cancer tissues and paracancerous tissues were collected from 60 gastric cancer patients and normal gastric tissues were collected from 30 normal people.DLC-1 m RNA level was determined by RT-PCR. DLC-1 protein in gastric carcinoma was detected and analyzed the relationship between DLC-1 expression and clinicopathological features of gastric cancer. Results Gastric carcinoma DLC-1 m RNA detection rate was 0.22±0.08 and it was significantly lower than that of adjacent tissues and(0.41±0.11)normal gastric tissue(0.63 ±0.13). In 60 cases of gastric cancer, the positive expression of DLC-1 protein was found in24 cases(60%).The positive expression rate of DCL-1 protein in patients with gastric cancer were not dependent on age,gender, and lesion size(P〈0.05); low differentiated gastric cancer patients with DLC-1 protein expression rate was25%. TNM stage III+IV patients DLC-1 protein expression rate was 29.17%, the depth of invasion in patients with T3 +T4 positive expression rate of DLC-1 was 33.33%, incidence of lymph node metastasis in patients with DLC-1 protein expression rate was 16.67%, the occurrence of gastric cancer patients with distant metastasis of DLC-1 protein expression rate of 20.83% was significantly lower than that of the corresponding high + differentiation in patients with stage I and II, TNM, T1+T2 in patients with invasive depth, no lymph node metastasis patients, distant metastasis free patients(P〈0.05). Conclusion The expression level of DLC-1 in the cancer tissue was significantly lower than those in the adjacent cancer tissues, and normal gastric tissues. DLC-1 protein and low differentiation, tumor stage, invasion depth, lymph node metastasis, and distant metastasis were correlated.
