摘要
目的探讨法舒地尔对大鼠低氧性肺动脉高压及肺组织中bFGF、VEGF表达的影响。方法雄性Wistar大鼠24只随机分为常氧对照组、低氧模型组、低氧法舒地尔干预组,检测各组平均肺动脉压力(mPAP)、右心室肥厚指数(RVHI);光镜下观察大鼠肺血管形态结构;RT-PCR检测bFGFmRNA、VEGFmRNA的变化。结果低氧模型组mPAP、RVHI值明显高于常氧对照组(P<0.01);法舒地尔干预组mPAP、RVHI显著低于低氧模型组(P<0.05)。光镜示,法舒地尔干预后大鼠的肺血管形态学改变较低氧组明显缓解。RT-PCR示,低氧模型组bFGF、VEGF值与其他两组比较明显升高(P<0.05)。结论法舒地尔对低氧所致的肺动脉高压、右心室肥厚及肺血管重塑疗效显著;bFGF、VEGF参与肺血管重塑的过程,其表达水平与肺血管重塑严重程度相关。
Objective To study the effect of fasudil on hypoxic pulmonary hypertension and the expression of bFGF and VEGF in lung tissue of hypoxic pulmonary hypertension in rats. Method Twenty-four male Wistar rats were di-vided randomly into 3 groups:normal control group,hypoxic model group and fasudil treatment group. We measured pulmonary arterial press and right ventricle hypertrophy index. The pulmonary artery morphological changes were ob-served under light microscope;basic fibroblast growth factor mRNA and Vascular endothelial growth factor mRNA were detectd by Reverse Transcriptase Polymerase Chain Reaction to the changes of. Result The level of mPAP and RVHI were significantlly higher in the hypoxic model group than those in normal control group and those of fasudil treated group(P〈0. 05). There were obviously morphological changes of pulmonary vascular that were alleviated in the fasudil treatment group than those in lower oxygen model group on Light microscopy. The mRNA expression of bFGF and VEGF were significantly increased than those in the the fasudil treatment group,and the normal control group (P〈0. 01). Conclusion Fasudil showed significantlly effective to prevent and reverse the development of pul-monary hypertension and right ventricular hypertrophy and pulmonary vascular from remodeling. bFGF and VEGF participate in the process of pulmonary vascular remodeling. The expression level of bFGF and VEGF relate directly to pulmonary vascular remodeling in severity.
