大鼠骨折骨痂中血管内皮生长因子的表达 被引量:1

Expression of Vascular Endothelial Growth Factor in Fracture Callus of Rats

摘要 目的探讨大鼠股骨骨折愈合中骨痂内血管内皮生长因子vascular endothelial growth factor,VEGF)的表达规律。方法取12周雌性SD大鼠32只,体重(280±25)g,随机分成4组,每组8只。A组为骨折1周组,B组为骨折2周组。C组为骨折3周组,D组为骨折4周组。制作股骨骨折模型。各组截取骨痂后行HE染色观察骨痂生长情况及其组织形态,免疫组织化学染色测定VEGF表达,原位杂交法测定VEGFmRNA表达。结果HE染色观察A组骨折间隙可见成纤维细胞,夹杂有少量的早期软骨细胞。B组有少量软骨骨痂形成。C组骨折端可见软骨骨痂增多和少量编织骨痂。D组骨折端编织骨痂增多,并出现板层骨。免疫组织化学和原位杂交方法检测可见术后1~4周的骨折部位,不同时间有多种组织细胞参与VEGF及VEGFmRNA的表达。骨痂中VEGF及VEGFmRNA平均阳性细胞百分率:创伤后7d骨折位点均表达增强,14d、21d仍保持高水平表达,28d骨折位点表达降低。结论VEGF在骨折修复过程中具有重要作用,并作为一种重要细胞因子参与和调节了骨折修复过程。 Objective To investigate the changes in the expression level of vascular endothelial growth factor (VEGF) in the bone callus of rats with femoral fracture. Methods Sixty- four 12 week - old SD rats weighing 280 + 25g were ran- domly divided into 4 groups (n = 8 each). The rats in Group A had a fracture for 1 week; the rats in Group B had a fracture for 2 weeks; the rats in Group C had a fracture for 3 weeks; and the rats in Group D had a fracture for 4 weeks. Fracture models of rat were established. Thus, the bone callus was obtained at 1 week, 2 weeks, 3 weeks, and 4 weeks after operation, respectively. Then, the bone callus growth and its histology were examined with the HE staining, the expression and changes in the level of VEGF were examined with the immunohistochemical staining, and the level of VEGF mRNA was measured with,the hybridization in situ. Results The HE staining indicated that some flboroblasts and early-stage chondrocytes were found in the fracture interspace in Group A, some cartilage callus in Group B, more cartilage callus and some woven callus in Group C, and more woven callus and some bone in Group D. VEGF and VEGF mRNA were detected in many kinds of cells in the fracture site during 1 week to 4 weeks after fracture by immunohistochemisty and hybridization in situ. The mean cell positive rates of VEGF and VEGF mRNA were as follow: an increase of VEGF protein expression around the fracture site was noted at 7th day, and increased at 14th and 21th days, which reduced at 28th day. Conclusions VEGF plays an important part in bone fracture repair and acts as an important cytokine participating and regulating fracture healing.
