摘要
目的 :探讨cyclinD1及 p2 1WAF1表达与大肠癌发生发展的关系。方法 :用免疫组化SP法对 40例大肠癌标本进行cyclinD1和 p2 1WAF1蛋白表达的检测 ,并与同期 2 8例大肠腺瘤、14例正常大肠粘膜作对比。结果 :cy clinD1和 p2 1WAF1的阳性表达率在大肠腺瘤 (92 .9% )和大肠癌 (6 7.5 % )均显著高于正常大肠粘膜 (35 .7% ) (P<0 .0 1,P <0 .0 5 )。cyclinD1的表达与大肠癌的分化程度及淋巴结转移无关。p2 1WAF1的阳性表达率在大肠癌 (37.5 % )显著低于正常大肠粘膜 (92 .8% ) (P <0 .0 1)。P2 1WAF1表达与大肠癌的分化程度及淋巴结转移有关。结论 :cyclinD1和 p2 1WAF1表达异常在大肠癌的发生、发展中是一个普遍事件。
To investigate expression of cyclinD 1 and p21 WAF1 and their relationship with multistage process of colon carcinogenesis.Methods:Immunohistochemical staining was performed with SP assay on archival materials from 40 colorectal carcinomas,28 colorectal adenomas,and 14 normal mucosa,cyclinD 1 and p21 WAF1 expression were compared.Results:The positive staining rate for cyclinD 1 in colorectal adenomas and carcinomas is 92.9% and 67.5% respectively,which are both higher than that in the normal mucosa (35.7%) (P adenoma <0.01,P carcinoma <0.05) and the expression of cyclinD 1 is not correlate with differentiation and lymph node metastasis of colorectal carcinoma.The positive stainimg rate for p21 WAF1 in normal mucosa (92.9%) is higher than that in colorectal carcinoma(37.5%) (P<0.01) and the expression of p21 WAF1 is correlate with differentiation and lymph node metastasis of colorectal carcinonma.Conclusions:The aberrant expression of cyclinD 1 and p21 WAF1 is a common event during multistage process of colon carcinogenesis.
