榆干离褶伞发酵液对血管内皮细胞的保护作用 被引量:10

摘要 为了探讨榆干离褶伞(Lyophyllum ulmarium,简写为LU)发酵液对人脐静脉内皮细胞(HUVEC)的保护作用,分别用20、40、80 mg/L LU干预HUVEC后用过氧化氢诱导凋亡,采用MTT法观察LU发酵液对细胞存活率的影响;通过比色法测定细胞上清液乳酸脱氢酶(LDH)活性;用流式细胞仪检测细胞凋亡率;Western blot法检测bcl-2、bax、Caspase-3、Caspase-9蛋白质的表达。结果显示:LU发酵液显著提高内皮细胞的存活率,降低细胞上清液LDH活性,抑制H2O2诱导的细胞凋亡,降低bax、Caspase-3、Caspase-9的表达,提高bcl-2的表达。这说明LU发酵液对血管内皮细胞有保护作用,其机制可能与抑制凋亡有关。 The protective effects of fermentation broth of Lyophyllum ulmarium (LU) on human umbilical vein endothelial cells(HUVEC) were observed.The H2O2 was used to induce the injury of HUVEC after the treatment with 20,40 and 80 mg/L LU.Survival rate of HUVECs was determined by the MTT assay,the lactate dehydrogenase(LDH) activity in culture serum was investigated using the colorimetric method,the apoptotic rate of HUVECs was detected by flow cytometry,and the protein expression of bcl-2,bax,Caspase-3 and Caspase-9 was detected by western blot method.The results showed that LU could increase the survival rate of endothelial cells induced by H2O2 and increase LDH activity,could reduce the cell apoptostic rate induced by H2O2,down-regulated the expression of bax,Caspase-3,Caspase-9 proteins,and up-regulated the expression of bcl-2 protein.It was indicated that the fermentation broth of Lyophyllum ulmarium has protective effects on vascular endothelial cell,probably via the inhibition of apoptosis.
