摘要
目的观察白藜芦醇对体外培养的病理性瘢痕成纤维细胞及TGF-β1/Smads信号通路的影响。方法不同浓度白藜芦醇处理病理性瘢痕成纤维细胞后,应用扫描电镜观察其形态学变化;MTT法检测其增殖状况;分别用反转录聚合酶链反应和免疫荧光细胞化学染色法检测其TGF-31/Smads信号通路中相关蛋白TGF-β1,Smads mRNA和蛋白的表达。结果病理性瘢痕成纤维细胞经白藜芦醇干预后,在形态学上可见到典型的凋亡特征;MTT法检测结果显示,各不同药物浓度组与对照组相比,OD值均偏低,差异有统计学意义(P<0.05);TGF-β1,Smad 2、3、4 mRNA和蛋白表达降低,而Smad 7 mRNA和蛋白表达增加,并且呈剂量依赖关系,与对照组相比,差异有统计学意义(P<0.05)。结论白藜芦醇能通过下调TGF-β1,Smad 2、3、4表达并上调Smad 7表达,来达到其抑制瘢痕组织成纤维细胞增殖并诱导其凋亡的作用。
Objective To observe the effects of resveratrol on fibroblasts of pathological scar and its TGF-β1/Smads signaling pathway. Methods After being treated with different concentrations of resveratrol, fibroblasts of pathological scar were observed by using scanning electron microscope; MTF method was used for the detection of the proliferation status;TGF-β1/Smads signal pathway associated proteins TGF-β1 ,Smads mRNA and protein expression were detected by polymerase chain reaction and immunofluoreseent cytochemical staining method. Results After the pathological scar fibroblasts were treated with different concentrations of resveratrol, the typical morphological features of apoptosis were seen. The results of MTT detection showed that the OD value of different concentration drug groups were lower as compared with that of the control group, there were significant differences ( P 〈 0.05 ). The TGF- β1, Smad 2,3,4 mRNA and protein expression increased,in a dose dependent relationship, in statistically significant (P 〈 0.05 ). Conclusion decreased, while Smad 7 mRNA and protein expression comparison with the control group, the difference was Resveratrol can down-regulate the expression of TGF- β1 ,Smad 2,3,4, and up-regulate the expression of Smad 7, to achieve the inhibition of scar tissue fibroblast proliferation and induction of apoptosis.
