摘要 目的采用响应面法优化龙眼核中总黄酮的提取工艺。方法在单因素试验的基础上,根据Box-Behnken试验设计原理,设计三因素三水平的响应面分析法,建立二次多项式回归方程的预测模型。结果各因素对黄酮提取率的影响顺序为:提取时间>丙酮浓度>提取温度。最佳提取工艺条件为:丙酮浓度59%,提取温度71℃,提取时间2.3 h。实际测得总黄酮平均提取率为43.67 mg/g,与预测值相符度97%。结论优选的工艺稳定可行,为工业生产提供了有利的借鉴,也为龙眼核的深入开发提供试验依据。 Objective To optimize the extraction conditions of total flavonoids from longan seeds by response surface analysis. Methods On the basis of single-factor experiment, the forecasting model was set up by using Box-Behnken design of three factors and three levels. Results The importance of the three factors affecting total flavonoid yield declined in the following sequence: extraction time, concentration of acetone and extraction temperature. The optimum extraction conditions were concluded as follows: acetone concentration of 59%, temperature of 71 ℃ and extractione time of 2.3 h. The measured extraction rate of total flavonoid was 43.67 mg/g, which was almost equal to the predicted value. Conclusion The optimized extraction process is steady and reliable, which could provide the reference for industrial production and resource exploitation of Dimocarpus longan seeds.
