摘要 目的建立密闭微波辅助-双水相萃取方法萃取香菇多糖方法并优化其萃取条件。方法采用单因素和正交试验方法考察了双水相组成、萃取温度、萃取时间、液料比等因素对香菇多糖萃取的影响。结果微波辅助-双水相萃取香菇多糖的优化条件为:硫酸铵质量百分数为19%,乙醇质量百分数为26%组成双水相,萃取温度为80℃,萃取时间30 min,液料比50∶1,经过验证香菇多糖的总平均萃取率为13.66%。结论微波辅助-双水相萃取香菇多糖方法具有快速,高效,选择性高的特点,可作为香菇多糖萃取和样品前处理方法。 OBJECTIVE To establish a novel method of pressurized microwave-assisted aqueous two-phase extraction( PMAATPE) of polysaccharides from Lentinula edodes,and to optimize its extraction condition. METHODS The influence of experimental factors including extraction temperature,extraction time,solvent-to-material ratio,ethanol concentration,ammonium sulfate concentration on the extraction of polysaccharides from Lentinula edodes were investigated by single-factor test and orthogonal tests.RESULTS The optimum condition of PMAATPE were aqueous two-phase system( ATPS) consisted of 19% ammonium sulfate and 26% ethanol,ratio solvent-to-material of 50 ∶1,extraction temperature of 80 ℃,and extraction time of 30 min. The total average yield of validation test for Lentinula edodes polysaccharides was up to 13.66%( w / w). CONCLUSION PMAATPE is a green,rapid and efficient alternative to extraction polysaccharides from Lentinula edodes with higher selectivity.
