摘要
目的为了萃取和分析山豆根多糖组成及其单糖组分的构成,建立高效液相色谱法(HPLC)测定山豆根多糖中的单糖组分的分析方法。方法利用微波辅助萃取结合三氟乙酸水解分别获得山豆根粗多糖及其水解产物单糖;采用1—苯基—3—甲基—5—吡唑啉酮(PMP)衍生化—HPLC法测定。结果所建立的PMP衍生化—HPLC法可准确地测定山豆根多糖中葡萄糖、甘露糖、鼠李糖、葡萄糖醛酸、氨基半乳糖、半乳糖和阿拉伯糖7种单糖组分,其线性范围、检测限、加标回收率和相对标准偏差分别为1.00-500μg/mL、0.101-0.213μg/mL、98.3%-102.3%和0.39%-1.3%之间。结果表明该多糖为这7种单糖组成的杂多糖,其摩尔比组成为:葡萄糖∶阿拉伯糖∶半乳糖∶甘露糖∶葡萄糖醛酸∶鼠李糖∶氨基半乳糖=60.0∶17.3∶6.69∶5.27∶4.25∶4.04∶2.45。
OBJECTIVE To investigate the composition of a polysaccharide extracted from Sophorae tonkinensis Gagnep.METHODS The polysaccharide were extracted and hydrolyzed by microwave assisted-extraction coupled trifluoro-acetic acid hydrolysis; monosaccharides were determined by 1-phenyl-3-methyl-5-pyrazolone( PMP) derivatization-high performance liquid chromatography( HPLC) with mobile phase of acetonitrile-0.05 mol / mL phosphate buffer solution( 18 ∶82,v / v) and detector at 245.0 nm. RESULTS Glucose,mannose,rhamnose,glucuronic acid,galactose,galactoseamine and arabinose in the polysaccharide extracted from the herb have been determined successfully by HPLC. Their liner ranges,detection limits,recoveries and RSD's are 1.00-500 μg / mL, 0. 101-0. 213 μg / mL, 98. 3%-102. 3%, and 0. 39%-1. 3%, respectively. The polysaccharide is a heteropolysaccharide composed of aforementioned seven monosaccharides,and its composition of the molar ratio is glucose ∶ arabinose ∶galactose ∶ mannose ∶ glucuronic acid ∶ rhamnose ∶ galactoseamine = 60.0 ∶17.3 ∶6.69 ∶5.27 ∶4.25 ∶4.04 ∶2.45.
