槲皮素对慢性肾衰大鼠肾脏细胞的保护作用研究 被引量:7

摘要 目的研究中药成分槲皮素对慢性肾衰(CRF)大鼠肾脏细胞损坏的保护作用。方法实验大鼠随机分为3组。CRF组和槲皮素治疗(QT)组以含腺嘌呤饲料喂饲8 w造成CRF;从第15 d开始分别经口给予生理盐水或槲皮素[100 mg/(kg·d)],共6 w;正常(NC)组未作干预。实验结束时,取存活大鼠肾制作病理切片,显微镜下观察计数损坏细胞和浸润细胞,进行统计分析。结果 NC组肾间质仅偶见浸润淋巴细胞(Lc)和多形核白细胞(PMN);CRF组浸润的Lc和PMN数大量增多;QT组浸润Lc和PMN数显著少于CRF组(P﹤0.01)。NC组肾小球和肾间质都未见空泡细胞,偶见破损细胞;CRF组肾小球及肾间质中空泡细胞和破损细胞显著高于NC组(P<0.01);QT组肾小球及肾间质中空泡细胞和破损细胞显著低于CRF组(P<0.01)。结论槲皮素对CRF大鼠肾间质炎性细胞浸润具有显著抑制作用,同时可以显著减少肾小球和肾间质实质细胞损坏。 Objective To study the protective effects of quercetin on renal cells of rats with chronic renal failure (CRF). Methods The selected rats were randomly divided into three groups. Feeds containing adenine were used in CRF and quercetin treatment (QT)groups in order to cause CRF. Since the 15th d, the two groups received the oral administration with saline or quercetin [ 100 mg/( kg ·d) 1 for 6 weeks. The normal group (NC) did not receive the intervention. At the end of the experiment, the kidneys of all the survival rats were collected and made into pathological sections. The defective and infiltrated cells were calculated and observed under the microscope and statistically analyzed. Results Few infiltrated lymphocytes ( Lc ) and polymorphonulear leukocyte ( PMN ) could be observed in the renal interstitium of NC group. The numbers of the infiltrated Lc and PMN in CRF group increased to a large amount ( 14.65 ± 2.36 and 10.34 ± 2.23 meanly in each field of high-powered optics). The number of Lc and PMN (8.68 ± 2.24 and 6.25 ± 1.58 ) in QT group was significantly lower than that in CRF group( P 〈 0.01 ). Physaliferous cells were not observed in the renal glomerulus and interstitium of NC group, and few defective cells could be seen. The percentages of physaliferous and defective cells in CRF group reached to 10.4 ± 2.2 and 22.6 ± 4.5, and the percentage of renal interstitium reached to 8.6 ± 2.2 and 23.4 ± 6, 4, which were all significantly higher than those in NC group(P 〈 0.01 ). The percentages of physaliferous and defective cells in QT group were 1.4 ± 0. 6 and 8.8 ± 2.6, respectively, and the percentage of renal interstitium was 1.2 ± 0.4 and 9.2 ± 3. 1, which were all significantly lower than those in CRF group( P 〈0.01 ). Conclusion Quercetin has obvious inhibitive effect on the infiltration of the inflammation cells in rats'renal interstitium and can also significantly reduce the damage to the parenchyma cells of renal glomerulus and interstit
