枸杞多糖对Aβ1~40诱导大鼠学习记忆功能减退的影响 被引量:2

摘要 目的:探讨枸杞多糖(LBP)对阿尔茨海默病(AD)大鼠学习记忆功能减退的影响。方法 SD健康大鼠随机分为5组:AD模型组,LBP高、中、低剂量干预组,空白对照组,每组24只。通过双侧海马注射Aβ1-40,建立AD大鼠模型。采用LBP连续灌胃6周作用于AD大鼠,观察各组大鼠行为学、海马SOD、GSH-Px活性和MDA含量以及BrdU标记阳性细胞表达的变化。结果(1)与AD模型组比较,LBP低、中、高剂量组潜伏期明显缩短、过平台次数明显增加(P〈0.05)。(2)与AD模型组比较,LBP低、中、高剂量组海马SOD、GSH-Px活性明显升高、MDA含量明显降低(P〈0.05)。(3)与AD模型组比较,LBP低、中、高剂量组齿状回BrdU阳性细胞表达明显增多(P〈0.05)。(4)LBP中、低剂量组比较,上述各指标均有明显差异(P〈0.05)。LBP高、中剂量组比较,上述各指标均无明显差异(P〉0.05)。结论 LBP可能通过提高海马SOD、GSH-Px活性、降低MDA含量以及增强齿状回BrdU阳性细胞表达,改善AD大鼠学习记忆功能。 Objective To investigate the effect of lycium barbarum polysaccharide (LBP) on Alzheimer's disease (AD)-induced learning and memory decline in rats. Methods The healthy SD rats were randomly divided into 5 groups:AD model group,low,medium and high-dose LBP treatment group,and control group,with 24 rats in each group. Aβ1~40 was injected into bilateral hippocampi to construct the AD rat model. The gastric lavage with LBP was performed for six consecutive weeks in AD rats. The variations of the behavior,the activities of antioxidant enzyme and malondiadehyde (MDA) content and expression of BrdU-labelled positive cells of each group were observed. Results (1)The latency was obviously shortened for low,medium and high-dose LBP treatment groups,while the number of crossing the platform was significantly increased,compared with the AD model group (P〈0.05).(2)The hippocampal SOD and GSH-Px activities were significantly increased and the MDA content decreased significantly for low,medium and high-dose LBP treatment groups (P〈0.05); compared with AD model group.(3)The expression of BrdU-labelled positive cells in all treatment groups were significantly increased,compared with the AD model group (P〈0.05).(4)All the indicators above showed significant variance between low-dose and medium-dose LBP groups(P〉0.05);but no significance in the indicators stated above was detected between high-dose and medium-dose LBP treatment groups (P〉0.05). Conclusion LBP relieves the AD-induced memory and learning decline in rats by enhancing the hippocampal SOD and GSH-Px activities,reducing the MDA content and promoting the BrdU-labelled positive cells in the dentate gyrus in the hippocampi.
