摘要
目的 :探讨松果体功能减退致褪黑素 (MT)分泌减少对 β-淀粉样蛋白 (Aβ)诱导海马细胞凋亡相关基因表达的影响。方法 :将 SD大鼠随机分两组 ,实验组摘除松果体 ,对照组给假手术 ,两组均在术后 40 d给 Aβ1~ 40 右侧海马注射 10 μg(10 μg/ μl) ,7d后标本用 SABC法检测凋亡相关基因 Bax,Bcl- 2、Fos蛋白表达。结果 :实验组 Aβ周围海马细胞凋亡相关基因 Bax(14.2 8± 4.18)和 Fos表达 (2 8.2 9± 6 .91)均比对照组 Bax(7.6 9± 2 .81)和 Fos表达 (0 .87± 0 .15 )增强 (P<0 .0 1) ;而 Bcl- 2在两组中表达 (7.48± 3.2 6 ,7.19± 3.37)无明显差别 (P>0 .0 5 )。结论 :褪黑素减少可加强
Objective:To investigate effects of decreased melatonin (MT) on the apoptotic related genes expression of hippocampal cells induced by amyloid βProtein (Aβ) following the pinealectomy. Methods: The rats were divided into two groups randomly. The experimental group was performed by pinealectomy, the control was made sham operation. Amyloid β protein 1~40(Aβ 1~40 )[10μg(10μg/μl)] was injected into the gippocalpus of the rats in the both groups above, at the fortieth days after operation. The protein expression of apoptotic related genes including Bax, Bal 2 and Fos in the brain tissues were immunohistochemically examined with SABC method at seventh day after injection. Results: The Bax and the Fos expression(14.28±4.18, 28.29±6.91 respectively) around Aβ in the hippocampal cells of the experimental group rats were more than the ones in the control (7.69±2.81, 0.87±0.15 respectively) ( P <0.01). The Bcl 2 protein expression, however, was no difference between the experimental (7.48±3.26) and the conterol group (7.19±3.37)( P >0.05)Conclusion: Decreasing melatonin following pinealectomy may enhance the tendency that amyloid β protein induced more apoptotic cells in the hippocampus of rats.
