摘要
以胞外多糖为评价指标,研究不同发酵条件对猪苓胞外多糖的影响。本试验采用了液体摇瓶培养的方法,在不同的温度、pH值、装液量和培养时间的条件下,测定猪苓发酵液中的胞外多糖,确定各单因素条件下猪苓胞外多糖的最佳发酵条件,再采用L9(34)正交试验设计优化其胞外多糖发酵条件,结果表明,猪苓胞外多糖发酵的最优条件为pH 5.0,装液量100mL,置于28℃的恒温培养箱培养9d,猪苓胞外多糖含量达到0.532g/100mL,说明通过优化发酵条件,可以有效提高猪苓菌丝体胞外多糖的分泌。
Effect of different fermentation conditions on extracellular polysaccharide of Polyporus umbellatus were studied according to the extracellular polysaccharide index.In this study,the liquid shake flask culture method was used.Under different temperatures,pH,fluid volume and incubation time,the content of extracellular polysaccharide of P.umbellatus in the fermentation liquor was determined to find the optimal fermentation condition in each single factor condition.Subsequently,the extracellular polysaccharide fermentation conditions were optimized by using L9(34) orthogonal experiment design,and the results showed that the optimal fermentation conditions for extracellular polysaccharide of P.umbellatus were:temperature 28℃,pH 5.0,incubation time 9 d and fluid volume 100 mL.Under the condition,the content of extraceilular polysaccharide ofP.umbellatus reached the maximum 0.532g/1 00mL,suggesting that optimization of fermentation conditions could effectively increase the secretion of extracellular polysaccharide of Grifola hyphae.
