摘要
目的 探讨胃癌组织中ＲａｓＰ２１血管内皮生长因子（Ｖａｓｃｕｌａｒ ｅｎｄｏｔｈｅｌｉａｌ ｇｒｏｗｔｈ ｆａｃｔｏｒ， ＶＥＧＦ）及微血管密度（Ｍｉ－ ｃｒｏｖｅｓｓｅｌ ｄｅｎｓｉｔｙ， ＭＶＤ）的关系。方法应用免疫组化Ｓ－Ｐ法检测４０例胃癌及２０例正常胃组织中ＶＥＧＦ、ＰａｓＰ２１及ＭＶＤ。结 果①正常胃组织中ＶＥＧＦ表达全部阴性：在胃癌组织中ＲａｓＰ２１、ＶＥＧＦ阳性分别为７０．０％、６２．５％。②ＲａｓＰ２１与ＭＶＤ及淋巴结 转移有显著差异（Ｐ＜０．０５）。ＶＥＧＦ的表达与细胞组织学分级、ＭＶＤ及淋巴结转移有显著差异（Ｐ＜０．０５）。在胃癌组织中ＲａｓＰ２１ 和ＶＥＧＦ的阳性表达强度与ＭＶＤ、淋巴结转移呈等级正相关（Ｐ＜０．０５）。③ＲａｓＰ２１的表达强度与ＶＥＧＦ表达强度呈等级正相 关。结论ｒａｓ基因可以通过增加ＶＥＧＦ产生引起瘤内ＭＶＤ增多，促进胃癌的生长和转移。检测ＲａｓＰ２１、ＶＥＧＦ表达强度，可以 作为判定胃癌转移潜能及评价预后的指标。
Objective To discuss the expression of VEGF, RasP21, and microvessel density (MVD) in gastric carcinoma and their relations. Methods We detected their expression in 40 cases of gastric carcinoma and 20 cases of the gastric tissues respectively by immunohistochemical S-P method. Results ①In normal gastric tissues, there was no expression of VEGF. In gastric carcinoma tissues, the expression of RasP21, and VEGF were 70.0 and 62.5 percent respectively. ②The expression of RasP21, had obvious difference with lymphnode metastasis and MVD (P<0.05). The expression of VEGF had obvious difference with lymphnode metastasis, MVD and histopathologic grade (P<0.05). The expressions of RasP21, and VEGF in gastric car- cionoma tissues positively correlated with MVD and lymphnode metastasis (P<0.05), but negahvely correlated with cyto- histopathologic grade (P<0.05). ③The expression of RasP21 and VEGF positively correlated. Conclusion The mutant of RasP21, can promte the growth and metastasis of tumor and increase the MVD in tumor. The detection of RasP21 and VEGF expression rate can be used as a speical method to determine the prognosis and metastasis in patient with gastric carcinoma.
