激光损伤视网膜后神经元凋亡及GFAP表达的研究

摘要 目的 观察兔视网膜激光损伤后神经元细胞有无凋亡 改变及视网膜Muller细胞胶质纤维酸性蛋白(GFAP)的表达变化。方法应用末端脱氧核苷酸转移酶介导的dUTP--X切口末端标记法(TUNEL法)标 记凋亡细胞。应用免疫组化染色显示视网膜Muller细胞GFAP表达。结果伤后6h、1、3、7d视网膜各层可见散在分布的TUNEL阳性凋亡 细胞,尤以外核层多见。伤后3d,视网膜可见Muller细胞GFAP表达;伤后14d,凋亡细胞数减少。随时间推移,GFAP表达增加。结论激光损伤诱发了视网膜神经元 的细胞凋亡,伤后Muller细胞GFAP表达增加与损伤修复有关。 Objective To determine the mechanism of retinal neuron death and the expression changes of GFAP by retinal Muller cells after laser injury.Methods A YAG laser was used to produce retina lesion in rabbits.Terminal deoxy nucleotidyl-transferase mediated dUTP and nick labelling(TUNEL)staining was used to identify the apoptotic neurons.The expression of GFAP by retinal Muller cells was located by immunohistochemistry.Results TUNEL positive apoptotic neurons were located in all the layers of retina in the 6th h our and on the 1st,3rd,and 7th day after injury .Apoptotic cells reduced in number on the 14th day after injury.The TUNEL positive cells were more densely located in the outer nuclear layer.The Muller cells expresse of GFAP starting to increase at 3 days after injury,and increased co ntinuously later.Conclusion Apoptosis is an import ant mechanism of retinal neuron death i n laser-injured retina,and the increased expression of GFAP by Muller cells is probably related to repair of the injury.
