摘要
2012年首次对杜鹃花科越桔属植物樟叶越桔原变种6’-O-咖啡酰熊果苷高产株的内转录间隔区(ITS)序列进行了PCR扩增和序列测定。通过BLAST同源搜索,采用邻位相连法构建了与其亲缘关系接近的越桔属8个种(或变种)的系统发育树,并分析了它们的亲缘关系。结果表明,参试材料与GenBank登记的长尾叶越桔(变种)和樟叶越桔ITS序列的同源性高达99%,显然属于种内变异水平。参试样本与长尾叶越桔(变种)和樟叶越桔的亲缘关系较近,遗传距离分别为0.008和0.010。研究结果将为寻找6’-O-咖啡酰熊果苷高产性状的近缘替代资源提供参考。
The internal transcribed spacer (ITS) region sequence analysis of 6' -O-caffeoylarbutin in high-yielding plant of Vaccinium dunalianum var. dunalianum (Ericaceae) was studied for the first time in 2012. Using the BLAST homology search in GenBank, the phylogenetic tree between Vaccinium dunalianum var. dunalianum and other Vaccinium species was constructed by neighbor-joining method, to discuss the genetic relationships of the se- lected species. The results showed the ITS sequences homology between the experimental material and V. dunalianum var. caudatifolium and V. dunalianum was 99 %. It was obviously belong to intraspecific varivtion. V. dunalianum var. dunalianum had the shortest genetic distance (0. 008 ) to V. dunalianum var. caudatifolium, and O. 010 of genetic distance to V. dunalianum. It provided a useful reference for finding substitutable resources with high-yielding of 6' -O-caffeoylarbutin.
