摘要
采用4种植物基因组DNA提取方法(CTAB法、SDS法、高盐法、试剂盒法)对八角基因组DNA进行提取试验。结果表明,对传统CTAB法稍加改良可以得到较为理想的提取效果,改良的内容包括①在加入CTAB提取缓冲液之前,先用CTAB-free缓冲液抽提1~2次;②将CTAB提取缓冲液中CTAB的用量从2%提高到3%;③提取缓冲液的用量由样品干重的10倍增加到60倍。用改良CTAB法提取的DNA经RAPD-PCR扩增,条带清晰,其质量能够满足分子标记的要求。
The genomic DNA of Illicium verum were extracted with 4 plant genomic DNA extraction methods,namely CTAB,SDS,high concentration salt and reagent kit.The results showed that the improved CTAB could achieve promising extraction effect.The modification included using CTAB-free buffer to wash out for 1~2 times prior to the procedure of adding CTAB extraction buffer;adjusting the used content of CTAB in extraction buffer from 2 % to 3 %;adding the extraction buffer to 60 times of the dry weight of leaf sample from 10 times.Using the DNA extracted by the improved CTAB method,through RAPD-PCR amplification,clear bands were achieved;and the quality of extracted DNA could sufficiently meet the requirements of further molecular marker experiment.
