摘要
目的:探讨血必净注射液影响巨噬细胞一氧化氮(NO)生成和白细胞介素(IL)-10与IL-12分泌的量效-时效关系。方法:体外培养小鼠RAW264.7巨噬细胞株,在无或有细菌脂多糖(LPS)诱导下,分别给予不同浓度(1、5、10 mg/ml)的血必净注射液干预,培养4、8和24 h后收集上清,分别采用Griess和ELISA方法检测NO、IL-10和IL-12p70的分泌水平。结果:与对照组比较,血必净注射液能够促进正常细胞IL-10的分泌,同时抑制NO和IL-12p70的分泌(P<0.05或P<0.01),这一效应能够持续到24 h,随着血必净浓度的增加,效应逐渐增强,其中高浓度抑制NO分泌能力最强,4 h和24 h较中浓度组有统计学意义(P<0.05或P<0.01),而中浓度促进IL-10分泌和抑制IL-12p70分泌的能力最强,中、高浓度组无统计学意义(P>0.05);LPS诱导自4 h便能够促进NO和IL-12p70的分泌(P<0.05或P<0.01),而8 h才能明显促进IL-10的分泌(P<0.01),随着时间延长,效应逐渐增强(P<0.05或P<0.01),血必净注射液能够抑制各时间点NO的分泌和促进IL-10的分泌,同时随着血必净浓度的增加,效应逐渐增强,其中高浓度效应最强,中、高浓度之间差异无统计学意义(P>0.05),中浓度血必净抑制IL-12p70的分泌效应最强,而中、高浓度间差异无统计学意义(P>0.05)。结论:血必净注射液能够影响小鼠巨噬细胞NO的分泌和IL-10与IL-12平衡,并存在明显的时效-量效关系。
Abstract Objective:To investigate the dose- and time-dependent effects of Xuebijing injection (血必净注射液) on the production of nitric oxide (NO) and interleukin (IL)-10/IL-12 in macrophages. Methods.. Murine RAW264.7 macrophage cell line was treated with different doses (1, 5 and 10 mg/mD of Xuebijing for various intervals (4, 8 and 24 hours) with or without lipopolysaccharide (LPS) stimulation. Cell-free supernatants were harvested to determine NO and IL-10/IL-12p70 levels by Griess analysis and ELISA kits, respectively. Results.. Compared to control group, treatment with Xuebijing could significantly enhance the secretion of IL-10, and also inhibit the secretion of NO and IL-12p70 in normal cells (P〈0. 05 or P〈0. 01). A high dose of Xuebijing showed the strongest inhibitory effect on the secretion of NO, and there were apparent differences between middle- and high-dose at 4 hours and 24 hours (P〈0. 05 or P〈0.01). However, a middle dose had strongest ability to promote IL-10 level and inhibit IL-12p70 level, and there were no marked differences in affect between middle- and high-dose of Xuebijing (P〉0.05). LPS stimulation to macrophages was shown to be able to significantly induce the secretions of NO and IL-12p70 after 4 hours (P〈0.05 or P〈0.01), but markedly increased IL-10 production after 8 hours (P〈0.01). Treatment with Xuebijing could inhibit the secretion of NO and increase the secretion of IL-10 in a dose-dependent response manner. Conclusion:The results suggest that Xuebijing injection can regulate the secretion of NO and IL-10/IL-12 balance in macrophages in a dose- and time-dependent manner.
