摘要
探讨微波辅助提取青果多糖的最佳条件。以硫酸-苯酚分光光度法(λ=490 nm)测定青果多糖含量,以青果多糖得率为评价指标,通过单因素试验和正交设计法优化提取工艺。最佳工艺:以水为萃取溶剂,固液比为1︰12(g/mL),微波功率500 W,提取温度为70℃,微波回流时间40 min/次,提取次数2次,多糖得率7.48%。此方法操作简单,提取效果较好。
The optimal extraction condition of the polysaccharide from Canarium album was studied with microwave method. The optimum extraction method of polysaccharide by microwave method from Canarium album was found with ingle-factor test and orthogonal experiments design. The content was determined by phenol-sulphuric acid spectrophotometry method (2=490 nm). The optimum microwave extraction conditions were microwave power 500 W, the ratio of material to water was 1 : 12 (g/mL), extracted times of 40 min each time, the microwave temperature of 70 ℃ and extracting 2 times. The content of Canarium album polysaccharides was 7.48% under this optimum extraction conditions. This method is easy to operate and obtain higher content ofpolysaccharide.
