多维液质联用系统分析养血清脑颗粒化学成分 被引量:12

Identification and characterization of constituents in Yangxue Qingnao Granules by multi-dimension HPLC-MS

摘要 目的采用高效液相色谱/飞行时间质谱(HPLC-TOF/MS)和高效液相色谱(二极管阵列)/电喷雾质谱(HPLC-DAD-ESI-MS/MS)的多维质谱联用系统,分析鉴定中药复方养血清脑颗粒(当归、川芎、白芍、熟地黄、钩藤、鸡血藤、夏枯草、决明子、珍珠母、延胡索、细辛)中化学成分。方法选用Agilent Zorbax C18(250 mm×4.6mm,5μm)色谱柱,以水(含0.1%甲酸)-乙腈为流动相梯度洗脱;电喷雾质谱在线检测,正离子和负离子模式下采集数据。结果通过比较保留时间、精确分子质量、多级质谱及PDA光谱,分析鉴定出养血清脑颗粒中41种成分。结论本方法从养血清脑颗粒鉴定出11种蒽醌、9种生物碱、11种酚酸、7种苯酞和3种单萜苷。 AIM To use a combination of HPLC-TOF/MS and HPLC-DAD-ESI-MS/MS to profile and identify Yangxue Qingnao Granules(Angelicae sinensis Radix,Chuanxiong Rhizoma,Paeoniae Radix alba,Rehmanniae Radix praeparata,Uncariae Ramulus cum Uncis,Spatholobi Caulis,Prunellae Spica,Cassiae Semen,Margaritifera Concha,Corydalis Rhizoma,Asari Herba).METHODS The chromatographic separation 50% methyl extract of tested granules was carried out on an Agilent Zorbax C18 column(250 mm×4.6 mm,5 μm).The mobile phase consisted of water-formic acid(100∶0.1,v/v)(A) and acetonitrile(B) with gradient elution.The analysis was performed in both negative and positive ion mode using full scan mode with ESI source.RESULTS Based on the retention time,mass spectra and UV spectra,forty-one compounds were identified or tentatively characterized.CONCLUSION The chemical constituents from Yangxue Qingnao Granules are identified as eleven kinds of anthraquinones,nine kinds of alkaloids,eleven kinds of phenolic acids,seven kinds of phthalides and three kinds of monoterpene glycosides.
