摘要
【摘要】目的探讨外阴鳞癌组织中E钙黏蛋白(E—cadherin)和13连环蛋白(p-eatenin)的表达及其意义。方法收集2005年1月至2012年4月在中国医科大学附属盛京医院妇产科行手术治疗的不同外阴病变患者的石蜡包埋标本63份,其中外阴鳞癌41份(外阴鳞癌组),外阴鳞状上皮内瘤变(VIN)Ⅱ~Ig22份(VINⅡ~Ⅲ组);以其中10份外阴鳞癌患者术后病理检查证实其切缘阴性的正常外阴组织标本作为对照组。采用免疫组化EliVision二步法检测3组标本中E—cadherin和β—catenin蛋白的表达情况,并对E—cadherin和β—catenin蛋白的表达与外阴鳞癌患者绝经状态、临床分期、病理分级及淋巴结转移的关系进行分析;采用Spearman相关分析法对外阴鳞癌、VINⅡ一Ⅲ组织中E.cadherin与β-eatenin蛋白的表达进行相关性分析。结果免疫组化EliVision二步法检测显示,外阴鳞癌组E—cadherin和β-catenin蛋白的异常表达率分别为46%(19/41)和61%(25/41),VINⅡ一Ⅲ组分别为64%(14/22)和68%(15/22),均明显高于对照组(均为0),差异均有统计学意义(P〈0.05);而外阴鳞癌组与VINⅡ~Ⅲ组比较,差异无统计学意义(P〉0.05);E—cadherin和β—catenin蛋白的异常表达率与外阴鳞癌患者的病理分级及淋巴结转移有关(P〈0.05),而与绝经状态及临床分期无关(P〉0.05)。Spearman相关分析显示,外阴鳞癌组41份组织中,E—cadherin和β-catenin蛋白均正常表达者14份,均异常表达者17份,两者的表达呈明显正相关关系(r=0.543,P=0.000);VINII~Ⅲ组22份组织中,E—eadherin和B.eatenin蛋白均正常表达者4份,均异常表达者11份,两者的表达无明显相关关系(r=0.295,P=0.182)。结论外阴鳞癌组织中E—cadherin和13-catenin蛋白存在异常表达,其异常表达与外阴鳞癌的病理分级及淋巴结转移相�
Objective To investigate the expression of E-cadherin and β-catenin in the Vulvar squamous cell carcinoma ( VSCC ) tissues. Methods A total of 63 documented paraffin blocks of VSCC (n = 41 ), vulvar intraepithelial neoplasia (n = 22), vulvar negative cutting edge tissues (n = 10) diagnosed in department of pathology of Shengjing Hospital of China Medical University from January 2005 to April 2012 were enrolled. EliVision immunohistochemical staining was used to detect the expression of E-eadherin and β-catenin in the three groups. Then to do a statistical analysis among the expression of them with patients' menopause status, pathological grade, clinical stage and lymph node metastasis. Spearman correlation analysis was used to analyse the expression of E-cadherin and β-catenin in the vulvar lesion tissues. Results The abnormal immunoreactivity for E-cadherin [ 46% ( 19/41 ), 64% ( 14/22 ) ] and β-catenin[61% (25/41),68% (15/22)1 in VSCC and VIN II - II1 were found, which were significantly different from that in normal epithelium samples ( P 〈 0. 05 ). The abnormal expression of E-cadherin and 15-catenin have no statistically significant difference between VSCC group and VIN Ⅱ - Ⅲ group (P 〉 O. 05). The abnormal expression of E-cadherin and β-catenin were collected with tumor pathological grade and lymph node metastasis status ( all P 〈 O. 05 ). The abnormal expression of E-cadherin and β-catenin have no statistically significant difference between menopause and the surgical stage of patients (all P 〉 0. 05). The abnormal expression of E-cadherin and [5-catenin have a significant positive correlation in the same sample in the VSCC tissue ( r = 0. 543, P = 0. 000 ) . The abnormal expression of E-cadherin and β-catenin have no correlation in the VIN Ⅱ - Ⅲ tissue ( r = 0. 295, P = 0. 182 ).
