摘要 目的探讨甲状腺正常组织和分化型甲状腺癌(DTC)中β-catenin和Cyclin D1的表达及临床意义。方法应用免疫组织化学链霉菌抗生素蛋白-过氧化物酶连接法(SP法)检测10 例腺瘤旁正常甲状腺组织和57例DTC标本中β-catenin和Cyclin D1的表达。结果β-catenin在甲状腺正常组织中均为正常表达,Cyelin D1均呈阴性。DTC中β-catenin和Cyclin D1的异常表达率分别为57.9%和64.9%。DTC中β-catenin异常表达与淋巴结转移密切相关;Cyclin D1表达与淋巴结转移和病理高分期有关。DTC中β-catenin和Cyclin D1表达呈明显正相关(P<0.01)。结论β-catenin异常表达可能激活靶基因Cyclin D1的表达,并在甲状腺癌发生发展中起到重要作用。 Objective To investigate the expression of β-catenin and Cyclin D1 in thyroid normal tissue and differentiated thyroid carcinoma (DTC), and their clinical significance. Methods The expression of β-catenin and Cyclin D1 in normal thyroid tissue around thyroid adenoma (10 cases) and DTC (57 cases) was detected by SP immunohistochemical technique. Results β-catenin was normally expressed in all of normal thyroid tissue, while Cyclin D1 wase negative. The aberrant expression rate of β-catenin and Cyclin D1 in DTC was 57.9 % and 64.9 % respectively. The aberrant expression of β-catenin was closely related to lymph node metastasis, and the expression of Cyclin D1 was related to lymph node metastasis and high TNM classification. The aberrant expression of β-catenin was significantly correlated with the expression of Cyclin D1 in DTC (P〈 0.01 ). Conclusion The aberrant expression of β-catenin may activate the expression of target gene Cyclin D1, which plays a pivotal role in the carcinogenesis and progression of thyroid carcinoma.
