摘要
目的:主要探讨姜黄素对人类宫颈癌Hela细胞中增殖作用的影响,以及细胞p53蛋白表达的影响。方法:本文通过不同浓度姜黄素干预人宫颈癌Hela细胞,采用MTT法检测药物对Hela细胞的增殖抑制率;RT-PCR和Western blot检测细胞凋亡相关蛋白p53表达。结果:MTT发现姜黄素对人宫颈癌Hela细胞有一定的抑制作用,并呈时间-剂量依赖性;RT-PCR结果显示p53m RNA的表达增加;western boltting显示p53蛋白的表达增加。结论:姜黄素可一直Hela细胞增殖,其机制可能是通过增加p53基因的表达而实现。
Objective: To observe the effect of cuicnmin on proliferation of human cervical cancer Hela cells and expression of p53 protein. Method: In this paper, Hela cells were treated with curcumin at various concentrations for various hours, Then determined for proliferation activity by MTT method, for transcription levels of β-catenin and p53 mRNAs by RT-PCR, and for expressions of β -catenin and p53 protein by Western blot. Result: Curcumin showed dose- and time-dependent inhibitory effect on the proliferation of Hela cells.Meanwhile, curcumin increased the mRNA transcription and protein expression levels of β -catenin and p53.Conclusion : Curcumin inhibited the proliferation of Hela ceils by a possible mechanism of increased the express of p53.
