姜黄素体外抑制人结肠癌癌细胞Caco-2增殖 被引量:5

摘要 目的研究姜黄素在体外对人结肠癌细胞Caco-2生长增殖及其抗肿瘤的相关分子机制。方法体外培养人结肠癌细胞Caco-2,四甲基偶氮唑盐(MTT)法检测姜黄素在相同浓度下、不同时间对于结肠癌细胞增殖的影响,并计算半数抑制量(IC50)值;反转录聚合酶链式反应(RT-PCR)法检环氧合酶-2(COX-2)、基质金属蛋白酶-9(MMP-9)的mRNA的表达影响。结果 MTT:相同干预浓度姜黄素抑制人结肠癌细胞增殖,其24 h,48 h,72 h的IC50分别为:205.737±14.929μmol/L、102.023±6.809μmol/L、86.627±9.005μmol/L;RT-PCR:人结肠癌细胞Caco-2正常对照组及姜黄素干预组MMP-9 mRNA灰度值分别为:0.786±0.101、0.310±0.046,P<0.05;COX-2 mRNA灰度值分别为0.830±0.173、0.063±0.035,(P<0.05)。结论姜黄素可能通过抑制COX-2及MMP-9的mRNA的表达来抑制结肠癌细胞Caco-2增殖。 Objective To investigate modulating effect of curcumin on the in vitro proliferation of human colon carcinoma cell line Caco - 2 and the molecular mechanism. Methods Ceils were incubated with curcumin in the same concentration for 24h, 48h, and 72h respectively. The cell viability was measured by MTT assay and IC50 was calculated. RT - PCR was used to evaluate the expression of COX - 2 and MMP - 9. Results The inhibitory effect showed a time - dependent manner; the ICs0 of the Caco - 2 cell line at 24h, 48h and 72h cultivation were 205. 737 ± 14. 929μmol/L, 102. 023 ±6. 809μmol/L, and 86. 627 ±9. 005 txmol/L, respectively. Analysis of RT - PCR product showed that the gray value of Caco - 2 MMP - 9 mRNA was 0. 786 ± 0. 101 in control group and 0. 310 ± 0. 046 in curcumin intervention group ( P 〈 0.05 ) ; the gray value of COX - 2 mRNA was O. 830 ± 0.173 in control group and 0. 063 ± 0. 035 in curcumin intervention group ( P 〈 0.05 ). Conclusion The antitumor mechanism of curcumin may be related to the inhibited Caco - 2 proliferation via COX - 2 and MMP - 9 mRNA expression inhibition.
