摘要
目的:探讨贵阳腐霉最佳的保存方法,以延长其有效保质期。方法:以菌丝体生长速度和灭蚊毒力为指标,观察分别在不同培养基、温度、氧分压条件下保存1、2、3、4、5个月的贵阳腐霉的质量,总结对该真菌保存影响较大的因素。结果:观察发现,在水琼脂培养基上保存的菌丝体生长速度最快;在葵花籽提取物(SFE)培养基上保存的菌丝体灭蚊毒力最高;4℃是贵阳腐霉长期保存的最佳温度,在-25℃和-75℃条件下菌丝体不能存活;氧分压分别为100%、66.7%、33.3%及0时保存的菌丝体差别不大,但33.3%氧分压保存的菌丝体稍好。结论:本研究总结了贵阳腐霉保存的最佳条件,为其商业开发和工厂化生产提供了有价值的资料。
Objective: To explore optimal conditions for long-term storage of Pythium guiyangense Su. Methods: Hyphal growth speed and virulence to Culex quinquefaciatus larvae were used as indexes to evaluate quality of P. guiyangense. Factors affecting storage of the fungus including media, temper- ature, partial pressure of oxygen, were tested as storage conditions or condition combination. The fungus was stored under different conditions and tested in 1, 2, 3, 4, and 5 months after storage initiation. Results: Mycelia stored on water agar grew the fastest, while that stored on SFE agar media had the highest virulence; fungus stored under 4 ℃ showed the best quality during the whole observation course, while that stored under -25 ℃ and -75 ℃ did not survive; Partial pressure of oxygen of 33.3% could meliorate the storage condition. Conclusions: The optimal storage condition combinations might provide valuable information for commercial development and industry production of P. guiyan- gense.
