摘要
目的探索贵阳腐霉最佳的保存方法。方法将贵阳腐霉保存在不同的温度、培养基和氧分压条件下,设计正交实验,分别保存2、4、6、8、10个月,观察基于这3个因素的最佳保存条件组合。结果综合分析各因素,长期保存后生长速度最快的组合是4℃-水琼脂培养基-33.3%氧分压;灭蚊毒力最佳的组合是4℃-SFE培养基-33.3%氧分压。在最佳条件组合下,贵阳腐霉可以保存10个月并保持正常生长活力,灭蚊毒力无明显下降。结论总结了保藏贵阳腐霉的最佳条件,为其以后的长期保藏提供实践依据。
Objective To explore optimal conditions for long-term storage of Pythium guiyangense Su. Methods Factors affecting storage of the fungus including temperature, media and partial pressure of oxygen, based on which orthogonal experiments were designed and carried out. The fungus was stored under different conditions and tested in 2, 4,6,8 and 10 months after storage. Relatively optimal conditions for P. guiyangense storage were summarized. Re- suits The optimal storage condition combinations were summarized,4 ~C-water agar-partial pressure of oxygen of 33.3% for better mycelial growth,and 4 ~C - SFE agar-partial pressure of oxygen of 33.3% for better virulence. Under the optimal conditions,P, guiyangense kept normal growth vigor and virulence after 10 months of storage, Conclusion The optimal storage condition combinations might provide practical substratum for long-term storage of P. guiyangense.
